Prevention of hypoglycemia by controlled relase of glucose from a solid dosage form

Abstract:

Tato diplomová práce se zabývá vývojem a reprodukovatelností výroby lékové formy k prevenci hypoglykémie, která vzniká jako nežádoucí efekt při terapii antidiabetiky, zejména inzulínem. Lékovou formu tvoří pelety s obsahem glukosy obalené nerozpustnou membránou, které uvolňují glukosu s časovou prodlevou. Pelety obsahují osmoticky aktivní látku, která způsobí rupturu obalové vrstvy, čímž dojde k řízenému uvolnění glukosy z pelet. Teoretická část práce se zabývá diabetem 1. i 2. typu, a také metabolismu glukosy v těle. V další část je věnována komplikaci vzniklé při konvenční léčbě insulinem- hypoglykémii. Následně je řešena otázka, proč jsou pelety nadějnou lékovou formou pro prevenci hypoglykémie a v závěru teoretické části práce se uvádí specifikace surovin a soupis použitých metod. Praktická část diplomové práce je věnována výrobě dvou složení pelet o třech šaržích. Tyto pelety obsahují vysoké procento glukosy jako účinnou látku a jsou potaženy speciální semipermeabilní membránou zajišťující řízené uvolňování glukosy. Tyto pelety jsou testovány, aby byla zjištěna reprodukovatelnost experimentu. V závěru práce je zjišťováno, v jakých intervalech dochází k uvolňování glukosy z pelet.

Abstract:

This diploma thesis focuses on production’s development and reproduction of a medication form for prevention of hypoglycemia which comes into existence as a side effect of an antidiabetic protocol, particularly insulin. The medication form consists of the pellets containing glucose and which are coated with insoluble membrane. Those pellest release glucose with a lag time. The pellets contain osmotically active excipient which creates rupture of coating layers which leads to the controlled release of glucose from the pellets. The theoretical part aims at diabetes type 1 and 2 and also at a metabolism of glucosy in a human body. The following part of the thesis is focused on a complication which arises while being on a convention insulin protocol – hypoglycemia. The aim of the subsequent part is to present the pellets as one of the auspicious treatments for prevention of hypoglycemia. There is a specification of raw material and a list of applied methods in the final section of the theoretical part. The practical part of the diploma thesis aims at a production of two pellet contents of three batches. Those pellets contain a high percentage of glucose as an active ingredient and they are coated with special semi-permeable membrane which provides the controlled release of glucose. The pellets are tested to find out the ability of reproduction of the experiment. in the end of the thesis, it is aimed at finding out the interval of the release of glucose.
