Evaluation of gene expression in selected tissue culter treated by microplastics

Abstract:

Diplomová práce se zabývá problematikou mikroplastů, jejich vznikem a následným uvolňováním do ekosystémů, což může vést k průchodu těchto částic do organismů. V organismech poté mohou mít mikroplasty toxické účinky a způsobovat změny fyziologických procesů. Detailněji jsou zde rozebrány známé vlivy na celou škálu druhů, a také je zde rozebrán výskyt mikroplastů v těle člověka. Zároveň byla popsána vazba látek na mikroplasty a s tím spojené změny účinků na organismy. V experimentální části byly sledovány změny exprese genů po ovlivnění mikročásticemi polystyrénu o velikosti 1 ?m a koncentraci 21 ?g/ml. Byly zjištěny změny exprese genů spojených s apoptózou a opravami DNA. U genů spojovaných s oxidačním stresem nebyla pozorována změna v případě experimentu v časové řadě, ale byla pozorována změna exprese superoxid dismutázy u vzorků s přidaným fetálním bovinním sérem a v jednom případě i ve vzorku bez séra, přičemž koncentrace byla opět 21 ?g/ml a doba ovlivnění 6 hodin. Při mikroskopování pomocí fluorescenčního mikroskopu Olympus BX53 bohužel nebylo možné rozlišit, zda mikroplasty procházely do buněk nebo docházelo pouze k jejich akumulaci v jejich okolí.

Abstract:

The thesis explores the issues caused by production of microplastics and its subsequent release to ecosystems. Furthermore, the thesis analyses the toxic effects and changes in physiological processes of microplastics on organisms. The known effects of microplastics on several species are examined in this thesis as well as the finding of microplastics within the human body. Additionally, the binding of substances to microplastics and associated effects of microplastics on the affected organisms were described in the thesis. The experimental part of the thesis investigates the changes in the gene after exposure of polystyrene microplastics with a size of 1 ?m and a concentration of 21 ?g/ml. Changes in the expression of genes associated with apoptosis and DNA repair were detected. For genes associated with oxidative stress no change was observed in the experimental case in the time series but a change in superoxide dismutase expression was observed in samples with added fetal bovine serum and in one case in a serum-free sample again at concentration of 21 ?g/ml and exposition for 6 hours. Using microscopy methods, it was not possible to distinguish if the microplastics passed into the cells or if they only accumulated in their surroundings.
