Analysis of telomere dynamics in vivo

Abstract:

Náplní této diplomové práce byla analýza dynamiky telomer a telomerových asociací in vivo pomocí fluorescenční mikroskopie živých buněk. Byly zavedeny a rutinně odzkoušeny stabilní a přechodné transfekce buněk a jako modelový materiál bylo použito osm buněčných linií, které se lišily mj. způsobem udržování telomer. Do buněk byly stabilně nebo přechodně vneseny expresní vektory pro fluorescenčně značené telomer-vazebné proteiny TRF1 a TRF2. Za účelem podrobných analýz dynamiky telomer a telomerových asociací byly vybrány tyto dvě nejvhodnější kombinace buněčné linie, vektoru a typu transfekce: a) buněčná linie U-2 OS transfekovaná stabilně vektorem pro telomer-vazebný protein TRF1 značený GFP a b) buněčná linie SK-LU-1 transfekovaná přechodně vektorem pro telomer-vazebný protein TRF2 značený EYFP. Vizualizované telomery byly detailně studovány pomocí vysokorozlišovací fluorescenční mikroskopie živých buněk v čase a získaná obrazová data byla analyzována ve 2D i 3D prostoru pomocí modulů programu ImageJ. Byly změřeny rychlosti jednotlivých telomer v buňkách U-2 OS včetně telomer asociujících a dále průměrné rychlosti všech analyzovaných telomer ve 2D i 3D prostoru. Potvrdilo se, že telomerové asociace jsou velmi dynamické struktury a jejich vznik a zánik dělí časové okamžiky v řádu několika minut. Tyto poznatky a metody budou dále využity pro detailnější analýzu pohybu telomer a telomerových asociací v buňkách s rozdílným mechanismem udržování délky telomer během buněčného cyklu a apoptózy.

Abstract:

The main goal of this diploma thesis was the analysis of dynamics of telomeres and their associations in vivo using fluorescent microscopy of living cells. We introduced and routinely conducted stable and transient transfections of cells and as a model material we used eight cell lines that differed in the mechanism of telomere maintenance. Expression vectors for fluorescently labeled telomere-binding proteins TRF1 and TRF2 were stably or transiently transfected into the cells. To analyze the dynamics of telomeres and their associations we selected two best combinations of cell lines, vectors and transfection types: a) cell line U-2 OS stably transfected with vector for telomere-binding protein TRF1 labeled with GFP and b) cell line SK-LU-1 transiently transfected with vector for telomere-binding protein TRF2 labeled with EYFP. Visualized telomeres were analyzed in great detail using time-lapse high-resolution fluorescent microscopy of living cells and obtained image data were analyzed in 2D and 3D by ImageJ software with special plug-ins. We measured the velocities of individual telomeres in U-2 OS cells including associating telomeres and we also measured the average velocity of all analyzed telomeres in 2D and 3D. Our results confirmed that telomere associations are very dynamic structures and the time interval from their formation to their disappearance is generally only several minutes long. These observations and methods will used in consequent analysis of dynamics of telomeres and their associations in cell lines.
