Effect of natural samples with different retinoid-like activity on embryonic development

Abstract:

Sinice jako jedny z nejrozšířenějších organismů na celém světě jsou schopny produkovat širokou škálu sekundárních metabolitů. Studie z poslední doby potvrzují, že do skupiny látek, produkovaných sinicemi, patří i látky s retinoidní aktivitou - retinoidy. Jelikož retinoidy jsou nepostradatelné při raných fázích vývoje obratlovců, existují obavy, že masivní rozvoj sinicových vodních květů může mít negativní dopad na jejich vývoj a životaschopnost. Ve své diplomové práci jsem se zaměřil na testování dvou standartních retinoidů (kyseliny all-trans retinové (atRA) a kyseliny 9-cis retinové (9-cisRA)) a dvou extraktů z biomasy přírodních vzorků sinicových květů in vivo s využitím embryonálního modelu s Danio rerio. Pro atRA byly hodnoty EC50 1,2 µg/l a hodnoty LC50 5,6 µg/l. To odpovídá výsledkům studií, které také testovaly atRA pro její teratogenní účinky (Selderslaghs et al. 2009; Selderslaghs et al. 2012; Wang et al. 2014; Jonas et al. 2015). Přírodní vzorky pocházely z oblasti Bítov, z vodní nádrže Vranov (Microcystis aeruginosa) s retinoidní aktivitou 6 650 ng atRA/g dw retinoidního ekvivalentu (REQ) a rybníku Ruda z oblasti Třeboň (Aphanizomenon klebahnii) s retinoidní aktivitou 4 400 ng atRA/g dw REQ. Oba testované extrakty způsobovaly stejné teratogenní efekty (deformace čelisti, ocasu, žloutkového vaku), které byly pozorovány i při testech s kyselinou all-trans retinovou. Po rozdělení vzorků do frakcí byly pozorovány teratogenní efekty jen v jedné frakci. Pozorované efekty ve frakcích s retinoidní aktivitou a v celém extraktu se vyskytovaly na statisticky významné hladině (p ≤ 0,05). Oproti tomu efekty ve frakcích bez retinoidní aktivity se na statisticky významné hladině nevyskytovaly (p ≤ 0,05). To dokládá hypotézu, že pozorované efekty způsobují retinoidní látky, které mohou sinice přirozeně produkovat do životního prostředí.

Abstract:

Cyanobacteria, as one of the most widespread organisms in the world, can produce a wide range of secondary metabolites. Recent studies have confirmed that substances with retinoid activity - retinoids - belong to the group of substances produced by cyanobacteria. Since retinoids are indispensable in the early stages of vertebrate development, there is concern that the massive development of cyanobacterial aquatic flora may have a negative impact on their development and viability. In my diploma thesis, I focused on testing two standard retinoids (atRA) of 9-cis retinoic acid (9-cisRA) and two extracts from biomass of natural samples of cyanobacteria in vivo using an embryonic model with Danio Rerio. For atRA, the EC50 values were 1.2 μg/l and the LC50 values were 5.6 μg/l. This corresponds to the results of studies that also tested atRA for its teratogenic effects (Selderslaghs et al., 2009, Selderslaghs et al., 2012, Wang et al., Jonas et al., 2015). Natural samples come from the Bítov area, from the Vranov water reservoir (Microcystis aeruginosa) with retinoid activity of 6,600 ng atRA/ g dw retinoid equivalent (REQ) and Aphanizomenon klebahnii Ruda fish with retinoid activity of 4,400 ng atRA/ g dw REQ. Both tested extracts caused the same teratogenic effects (jaw deformation, tail deformation, and yolk sac deformation), which were also observed with all-trans retinoic acid testing. After fractionation of samples into fractions, teratogenic effects were observed in only one fraction. The observed effects in the fractions with retinoid activity and whole extract were statistically significant (p ≤ 0.05). In contrast, the effects in non-retinoid fractions were not statistically significant (p ≤ 0.05). That support hypothesis that the observed effects cause retinoids that can naturally produce cyanobacteria in the environment.
