Live vaccines, innate immune response and chicken colonisation with Salmonella

Abstract:

Oslabené kmeny Salmonella enterica lze využít pro expresi heterologních antigenů. Schopnost sekrece proteinů však u atenuovaných kmenů salmonel nebyla studována. V první části své práce jsem proto porovnávala proteinovou sekreci u mutantů rfaL, rfaG, rfaC, lon, htrA, clpP, aroA, phoP a spoT-relA ve kmenech S. enterica serovar Typhimurium a Enteritidis. Defekt v sekreci proteinů byl zaznamenán u mutantů rfaG, rfaC, lon a spoT-relA, které jsou tudíž nevhodné jako vektory pro expresi a sekreci heterologních antigenů. Podrobněji jsem se zabývala mutanty S. Enteritidis a Typhimurium rfaC, rfaG a rfaL s defektem v syntéze lipopolysacharidu (LPS). U všech tří rfa mutantů byla zjištěna narušená motilita na polotuhém LB agaru, ačkoliv rfaL mutant secernoval bičíkový protein FliC. Mutantní kmeny rfaC a rfaG žádné extracelulární proteiny, včetně flagelinu, nesecernovaly. U rfaC mutanta S. Typhimurium však docházelo k expresi FliC, detekovanému v cytoplazmě pomocí western blot analýzy, ve stejném množství jako u divokého kmene. Oligosacharidové jádro LPS je nezbytné pro sestavení struktury bičíku a kompletní O-antigen LPS je potřebný pro správnou funkci bičíku. Striktní hygienická opatření v chovech drůbeže přinášejí kromě nesporných výhod i možné problémy. Kuřata v čistých líhních nepřicházejí do kontaktu s rodičovskou mikroflórou a po vylíhnutí mohou být kolonizována náhodnými bakteriemi z prostředí. V druhé části práce jsem proto sledovala vývoj střevní mikroflóry, imunitní odpověď na kolonizaci střeva a infekci Salmonella Enteritidis u kuřat různého věku. Komplexnost střevní mikroflóry u kuřat postupně narůstala od 1. do 19. dne života. Během prvních 3 dnů po vylíhnutí bylo střevo kuřat chráněno zvýšenou expresí β-defensinů, poté jejich exprese klesla a 4. den života byla nahrazena maximální expresí cytokinů IL-8 a IL-17. Ve věku 4 dnů docházelo u kuřat k přechodnému, mírnému, fyziologickému zánětu a tím i k vyzrávání imunitního systému střeva, které se projevilo přechodem z Th-1 imunitní odpovědi u 1- a 4-denních kuřat infikovaných S. Enteritidis (charakterizovanou indukcí IFN-γ a iNOS) na Th-17 imunitní odpověď u 16-denních kuřat infikovaných S. Enteritidis (charakterizovanou indukcí IL-17 a IL-22). Zrání imunitního systému střeva tak zvyšuje rezistenci kuřat k infekci salmonelami a pravděpodobně i jinými patogeny.

Abstract:

Attenuated strains of Salmonella enterica are frequently used as a vector for the expression of heterologous antigens. However, the ability of protein secretion in attenuated Salmonella strains has not been studied. In the first part of the study I have therefore compared protein secretion of S. enterica serovar Enteritidis and Typhimurium rfaL, rfaG, rfaC, lon, htrA, clpP, aroA, phoP and spoT-relA mutant strains. Defective protein secretion was detected in rfaG, rfaC, lon and spoT-relA mutants which are therefore unsuitable as vectors for the expression and secretion of heterologous antigen. S. Enteritidis and Typhimurium rfaC, rfaG and rfaL mutant strains with defects in lipopolysaccharide (LPS) synthesis were characterised in detail. All three rfa mutants were defective in motility in semisolid LB agar although rfaL mutant secreted flagellar protein FliC. Both rfaC and rfaG mutant strains did not secrete any extracellular proteins, including flagellin. However, S. Typhimurium rfaC mutant expressed FliC protein, which was detected by western blot analysis in cell cytoplasm in the same amount as in wild type strain. Oligosaccharide core of LPS is therefore essential for the flagellar structure assembly and complete LPS O-antigen is required for correct flagellar function. Strict hygienic standards in poultry farming cause besides indisputable benefits also possible problems. Chickens hatched in clean hatcheries will never get into contact with microbiota of parents and can be colonized with random environmental bacterial species. In the second part of the study I have therefore characterised gut microbiota development and immune response to natural microbiota colonization and to Salmonella Enteritidis infection in chickens of different age. The complexity of chicken gut microflora gradually increased from day 1 to day 19 of chicken life. During the first 3 days of life, chicken cecum was protected by an increased expression of β-defensins expression of which decreased since day 4. Instead, a transient, moderate physiological inflammation leading to the gut immune system maturation was recorded on day 4 of chicken life. This led to a switch from Th-1 immune response of 1- and 4-days old chickens infected with S. Enteritidis (characterized by induction of IFN-γ and iNOS) to Th-17 immune response of 16-days old S. Enteritidis infected chickens. Maturation of the gut immune system then increased resistance of chickens to infection with Salmonella and likely also other pathogens.
