Abstract:

Auxin, ve své nejběžnější formě kyselina indol-3-octová, je esenciální rostlinný hormon s významnou rolí v takřka všech stádiích rostlinného vývoje. Ovlivňuje dělení buněk a jejich prodlužovací růst (Rayle & Cleland, 1970; Evans et al., 1994), embryogenezi (Benková et al., 2003; Friml et al., 2003), formování meristémů (Benková et al., 2003; Bainbridge et al., 2008; reviewed in Yamoune et al., 2021), tvorbu cévních pletiv (Scarpella et al., 2006; Bishopp et al., 2011), gravi- a fototropismus a mnoho dalších morfogenetických procesů. Auxin je dle klasických schémat syntetizován převážně ve vrcholovém meristému, například v mladých listech či pupenech (Teale et al., 2006; Stepanova et al., 2008). Z místa syntézy do místa působení je auxin přenášen koordinovanou činností přenašečů auxinu lokalizovaných polárně na plazmatické membráně buněk. Navzdory značnému badatelskému úsilí zůstávají stále mnohé detaily spojené s přenosem auxinu, jejž zprostředkovává protein PIN, nezodpovězeny. Alternativní sestřih (AS) je zásadním prvkem regulace genové exprese, který významným způsobem zvyšuje různorodost genové exprese. Poměr hladin jednotlivých transkriptů může být v různých tkáních i za specifických podmínek různý. Tím může potenciálně nastavit odpovídající vývojový program vzhledem k měnícím se vnějším podmínkám. Navzdory všemu jsou rozsah a role AS ve vývoji rostlin, včetně procesů na auxinu závislých, poměrně málo znám. V článcích, které jsou základem této dizertace, představuji AS jako relevantní regulátor transportu auxinu. Zde popisuji dvě sestřihové izoformy proteinu PIN7, kritického auxinového přenašeče. Ve své práci jsem prozkoumal jejich molekulární vlastnosti a způsob jejich vzájemné interakce v kontextu současného vědění o polárním transportu auxinu. Navržený molekulární model jsem ověřil rozsáhlými fenotypovými analýzami, metodami konfokální mikroskopie, ověřováním protein-proteinových interakcí a přímým měřením transportu auxinu. Na základě výše uvedeného jsem popsal úzké propojení mezi laterální mobilitou auxinových transportérů na plazmatické membráně a jejich schopností transportovat auxin. Rovněž spolu se spoluautory popisujeme vývoj reportérového systému in vivo, který umožňuje vizualizaci sestřihu PIN7 v pletivech Arabidopsis. Zjistili jsme, že po přidání auxinu dochází k nápadné změně exprese jednotlivých isoforem PIN7. V neposlední řadě jsme vyvinuli systém pro identifikaci faktorů, které ovlivňují expresi PIN7 pomocí genetického screeningu. Tento systém je založený na myšlenkovém principu využitém v popsaném fluorescenčním reportérovém systému. V neposlední řadě jsem spolu s kolegy shromáždil nejvýznamnější příklady AS objevených v rostlinách, které jsme zcela nově uspořádali dle základních molekulárních interakcí, které byly mezi sestřihovými izoformami doposud dokumentovány.

Abstract:

Auxin, in its most common form indole-3-acetic acid (IAA), is an essential plant hormone that plays a role in nearly all steps of plant development. It controls cell division and elongation (Rayle & Cleland, 1970; Evans et al., 1994), embryo development (Benková et al., 2003; Friml et al., 2003), meristem maintenance (Benková et al., 2003; Bainbridge et al., 2008; Yamoune et al., 2021), vascular tissue formation (Scarpella et al., 2006; Bishopp et al., 2011), gravi- and phototropism, and many other morphogenetic processes. Auxin is mostly synthesized in the shoot apical meristems e.g., young leaves, flower buds (Teale et al., 2006; Stepanova et al., 2008). From the sites of biosynthesis auxin is delivered to the effector tissues by the coordinated action of polar auxin transporters. Auxin fluxes are finely regulated by the auxin transporters from the PIN-formed (PIN) family. Despite previous tremendous research effort, many details about the PIN-mediated auxin transport remain unanswered. Alternative splicing (AS) is an essential element in the gene expression networks that significantly increases transcript and protein diversity. The ratio and pattern of AS can vary in different tissues or under specific stimuli which has a potential to set appropriate developmental programs and/or adapt to the changing environmental conditions. However, the extent and mechanistic role of AS in plant development, including in auxin-related processes, remain poorly described. In the set of studies summarized in this thesis, I introduce AS as a relevant regulator of auxin transport on the example of PIN7, central auxin transporter. In the article, comprising the core of this thesis, I have collaboratively characterized two PIN7 splicing isoforms, examining their molecular properties and mode of mutual interaction in the context of the current knowledge about the polar auxin transport. I supported the identified molecular model with extensive phenotypic analyses, confocal microscopy, protein interaction assays, and auxin transport measurements. Hence, I propose a tight connection between the lateral mobility of PIN7 protein isoforms in the plasma membrane and their auxin transport capabilities. In the accompanying article, together with the collaborators I described an in vivo system of reporters allowing for the tissue-specific visualization of AS of PIN7. I also showed that PIN7 isoform expression is changed following the exogenous auxin application. Ultimately, based on the fluorescent reporter, we developed a system for genetic screening for the factors upstream of the PIN7 AS event. Additionally, in the review article, I and colleagues gathered the best-known examples of the protein-productive AS and summarized them to underline the common modes of action and functions of resulting protein isoforms.
