Determination of alkylphenols in environmental matrices

Abstract:

Předkládaná disertační práce se zabývá stanovením alkylfenolů v environmentálních matricích jako je voda a půda. Pozornost byla věnována především průmyslově nejrozšířenějším analytům, jako jsou: 4-terc-oktylfenol (4-t-OP), 4-oktylfenol (4-OP), 4-n-nonylfenol (4-n-NP), isomery 4-nonylfenolu (iso-NP) nazývány také jako technický nonylfenol, bisfenol A (BPA) a bisfenol F (BPF). Náplní práce bylo optimalizovat a validovat analytickou metodu pro stanovení výše uvedených alkylfenolů a bisfenolů v environmentálních matricích (voda, půda) s vyuţitím 5-(dimethylamino) naftalen-1-sulfonyl chloridu (dansyl chlorid, DNSC) jako derivatizačního činidla. Pro separaci a kvantifikaci dansyl derivátů alkylfenolů byla pouţita metoda vysokoúčinné kapalinové chromatografie s tandemovým hmotnostně spektrometrickým detektorem, přičemţ ionizace probíhala s vyuţitím elektrospreje v pozitivním módu (HPLC-ESI+/MS/MS) (Článek I). Pro půdu jako komplexnější matrici byly testovány čtyři extrakční techniky jakými byly Soxhletova extrakce, extrakce pomocí ultrazvuku (USE), urychlená extrakce rozpouštědlem (ASE) a rychlá, snadná, levná, efektivní, robustní a bezpečná extrakce nazývaná QuEChERS. QuEChERS technika má mnoho modifikací jako je originální, citrátová a acetátová, které byly také testovány pro vybrané půdy v závislosti na celkovém organickém uhlíku (TOC) (Článek II). Analytické metody byly aplikovány na reálné vzorky pitné vody z vodovodního potrubí (Článek III), sladkovodní rybníky a nádrţe ovlivněné vodním květem, u kterých byla měřena estrogenní aktivita a koncentrace estrogenních látek (Článek IV). Dalšími reálnými vzorky byla balená pitná voda v PET lahvích, voda z čističky odpadních vod, artificiální půdy a sada půd vybraná z banky půd Centra pro výzkum toxických látek v prostředí (RECETOX). Dále byly realizovány experimenty týkající se analýzy blanku, jako notoricky známý problém v ultrastopové analýze těchto analytů (Článek V). Publikované práce, které jsou součástí předkládané disertační práce (Články I–V) obsahují detailní popis dané problematiky.

Abstract:

The presented thesis deals with the determination of alkylphenols in environmental matrices, water and soil. Attention was devoted mainly to the industrially most widespread analytes such as 4-tert-octylphenol (4-t-OP), 4-octylphenol (4-OP), 4-n-nonylphenol (4-n-NP) isomers of 4-nonylphenol (iso-NP) called also technical nonylphenol, bisphenol A (BPA) and bisphenol F (BPF). The aim of the thesis was to optimize, validate and use analytical methods for the determination of the above-mentioned alkylphenols and bisphenols in environmental matrices (water and soil) applying 5- (dimethylamino) naphthalene-1-sulfonyl chloride (dansyl chloride, DNSC) as a derivatizing agent. For the separation and quantification of the dansyl derivatives, the high-performance liquid chromatography method with a tandem mass spectrometer detector was used. The ionization was performed using the positive mode of electrospray (HPLC-ESI+/MS/MS) (Article I). Soxhlet extraction, ultrasound extraction (USE), accelerated solvent extraction (ASE) and fast, easy, cheap, efficient, robust and safe extraction (QuEChERS) were tested for soil as a more complex matrix. The QuEChERS technique has several modifications such as original, citrate and acetate, which have also been tested for selected soils based on total organic carbon (TOC) (Article II). The analytical methods were applied to real samples of drinking water from the water pipeline (Article III) as well as freshwater ponds and reservoirs affected by water blooms where estrogenic activity and estrogenic concentration were determined (Article IV). Other real samples were bottled drinking water in PET bottles, sewage water, artificial soil, and real soils selected from the Soil Bank from the RECETOX Research Center. Additionally, experiments were conducted on the analysis of the blanks as a notorious problem in ultra-trace analysis of alkylphenols (Article V). The published papers which are an integral part of the presented thesis (Articles I–V) give a detailed description of the issue.
