Abstract:
Bakalářská práce se zabývá in vitro syntézou tRNA. Při této technice se využívá jednoduchosti transkripce bakteriálních systémů, které narozdíl od eukaryot neobsahují komplikovaný mechanismus řízený transkripčními faktory. V této práci byla přepsána sekvence odpovídající iniciátorové tRNA pro methionin, která neobsahuje modifikace potřebné k svému správnému sbalení.Tento substrát bude dále použit pro …more
Abstract:
The theme of this thesis is In vitro synthesis of tRNAs. The technique uses simple bacterial systems of transcription which contrary to eukaryotes doesn’t need transcription factors to control the transcription process. In this work the iniciator tRNA(Met) which lacks specific modification for proper folding has been transcribed. Synthesized tRNA will be used for further study of RNA quality control …more
