Study of proteins associated with congenital anemia

Abstract:

Vrozená anémie je skupina vzácných dědičných onemocnění, která se vyznačují neefektivní erytropoézou. Typickými fenotypovými znaky vrozené anémie typu I (CDA-I) jsou mezijaderné můstky a houbovitý heterochromatin v erytroblastech. Nemoc CDA-I je spojená s mutacemi dvou proteinů – Codanin1 a CDIN1. Codanin1 reguluje přísun histonů do jádra během S-fáze buněčného cyklu – je tedy esenciální pro organizaci chromatinu a zachování primární funkce genomu. CDIN1 byl objeven teprve nedávno a jeho funkce dosud nebyla jednoznačně určena. Dosavadní experimentální výsledky však ukazují, že CDIN1 váže C-konec proteinu Codanin1, a naznačují, že se oba proteiny účastní společné regulační dráhy. Na základě nejnovějších výsledků se předpokládá, že patogenní mutace proteinu CDIN1 rozrušují interakci s proteinem Codanin1, což vede k rozvoji nemoci CDA-I. V rámci této práce byly připraveny a zkoumány mutantní varianty proteinu CDIN1 se záměnou aminokyselin – Y94S a L178Q – in vitro. Kromě toho byly zjišťovány biofyzikální charakteristiky varianty CDIN1 s mutací L178Q. Výsledky experimentů naznačují, že mutace Y94S a L178Q snižují stabilitu proteinu CDIN1. Závěrem následných interakčních analýz bylo, že mutace Y94S a L178Q proteinu CDIN1 narušují jeho vazbu na Codanin1.

Abstract:

Congenital dyserythropoietic anaemia (CDA) is a group of rare hereditary diseases characterised by ineffective erythropoiesis. Typical features of CDA type I include internuclear bridges and spongy heterochromatin in erythroblasts. CDA-I is associated with mutations in two proteins—Codanin1 and CDIN1. Codanin1 acts as a negative regulator of histone supply during the S-phase of the cell cycle, which makes it importnant for chromatin organization and maintaining genome function. CDIN1, however, has been discovered recently and its function has not been fully described. Nevertheless, experimental data demonstrate that the protein CDIN1 binds to the C-terminus of Codanin1 and predict participation in the same regulatory pathway. Recent findings suggest that CDIN1 pathogenic mutations disrupt the essential CDIN1-Codanin1 interaction, therefore, result in CDA-I development. In this thesis, two mutated CDIN1 variants associated with CDA-I—Y94S and L178Q—were prepared and tested in vitro. Additionally, CDIN1 L178Q was biophysically characterized. All gathered experimental data indicate that said pathogenic mutations destabilize CDIN1. Subsequent interaction analyses revealed that the Y94S and L178Q mutations of CDIN1 disrupt its binding to Codanin1.
