Characterization of biomolecules using the mass spectrometry and separation techniques

Abstract:

Předkládaná disertační práce je zaměřena na studium oligomerů s využitím hmotnostní spektrometrie a separačních technik. Disertační práce je koncipována jako komentovaný soubor prací již publikovaných. Každá práce je opatřena stručným komentářem dosažených výsledků. Práce je složena ze tří ucelených studií. První studie se zabývá využitím desorpce a ionizace laserem pomocí matrice (MALDI) ve spojení s průletovým hmotnostním spektrometrem (TOF MS) s minimální předúpravou pro kvalitativní analýzu fruktooligosacharidů ve střevech selat. Výsledky jsou srovnány s analýzou na tenké vrstvě. Ve druhé studii je rozpracována metoda kvantifikace fruktooligosacharidů pomocí MALDI TOF MS bez použití izotopového značení. Ke kompenzaci nelineární MALDI odezvy jsou použity umělé neuronové sítě. Součástí studie je i optimalizace extrakce fruktooligosacharidů z reálné matrice pro MALDI TOF MS analýzu. Třetí studie se zabývá analýzou oligoargininů pomocí spojení HPLC s hmotnostní spektrometrií a tandemovou hmotnostní spektrometrií. K těmto účelům je použita modifikace guanidinové skupiny argininu malondialdehydem. Optimalizovaná metoda je použita pro sledování čistoty dendritických peptidů obsahující oligoargininy v bočním řetězci. Poslední studie doplňuje disertační práci o další možnosti analýzy pomocí MALDI TOF MS. Studie se zabývá charakterizací abietinové kyseliny a giberelinové kyseliny na různých chromatografických rozhraních.

Abstract:

This dissertation thesis is focused on study of oligomers using mass spectrometry and separation techniques. This thesis is conceived as a series of published papers. Results of all papers are commented and discussed. The work includes three studies. The first study concerns application of matrix assisted laser desorption ionization (MALDI) coupled with time of flight mass spectrometry (TOF MS) for analysis fructooligosaccharides in the intestine of piglets. The results are compared with thin layer analysis. In the second study, a method for MS quantification of fructooligosaccharides using MALDI TOF MS without isotopic labelling is developed. To compensate nonlinear MALDI response, artificial neural network is applied. This study includes optimalization extraction techniques for fructooligosaccharides in food supplements. The third study deals with the analysis oligoarginines using HPLC combined with mass spectrometry and MS/MS. For this purpose, malodialdehyde modification of the guanine group is used. The optimized method helps to identify major side products of dendrimeric peptide. Recent study completes disseration thesis and posibility MALDI TOF MS analysis. Recent study characterizes abietic and gibberellic acid on different chromatografic substrates.
