Combination Therapy: A Synergistic Strategy Against Bacterial Infections

Abstract:

Výskyt antibiotické rezistence se globálně zvyšuje rapidním tempem. Bez naléhavých opatření a vývoje nových antimikrobiálních látek, směřujeme do postantibiotické éry, ve které mohou běžné infekce a drobná poranění zabíjet. Proto je důležité se zabývat vývojem nových antimikrobiálních činidel, která budou účinnější než dosavadní antibiotika. Předložená disertační práce se zabývá alternativní strategií v léčbě bakteriálních infekcí na bázi kovů. Byl zkoumán účinek kombinované terapie komplexu železa s ampicilinem vůči Grampozitivní bakterii Staphylococcus aureus na fenotypové a genotypové úrovni v porovnání se samotným ampicilinem. Další navrženou strategií bylo využití pokročilých robotických materiálů s inkorporovaným antimikrobiálním peptidem indolicidinem a jejich účinek byl stanoven na biofilmu methicilin-rezistetní bakterii S. aureus (MRSA). U obou studií byl zjištěn synergický a inhibiční efekt testovaných látek vůči jednotlivým testovaným bakteriím.

Abstract:

Antibiotic resistance is increasing at a rapid pace globally. Without urgent action and the development of new antimicrobial agents, we are heading into a post-antibiotic era in which common infections and minor injuries can kill. Therefore, it is important to deal with the development of new antimicrobial agents that will be more effective than existing antibiotics. The submitted dissertation deals with an alternative strategy in the treatment of metal-based bacterial infections. The effect of combined therapy of an iron complex with ampicillin against the Gram-positive bacteria Staphylococcus aureus at the phenotypic and genotypic level was investigated in comparison with ampicillin alone. Another proposed strategy was the effect of advanced robotic materials incorporating the antimicrobial peptide indolicidin and determined on methicillin-resistant S. aureus (MRSA) biofilm. In both studies, a synergistic and inhibitory effect of the tested substances against individual tested bacteria was found.
