Effects of pre-sowing seed treatment on occurrence of pathogens from genus Tilletia

Abstract:

Tato práce se zabývá účinností předseťové úpravy osiva v návaznosti na výskyt patogenů přenosných osivem, se zaměřením na rod Tilletia. V teoretické části byla popsána problematika moření. Zmíněny byly okolnosti správného výběru mořidla, kvalita moření a její testování, byly nastíněny také možnosti fyzikálních metod ošetření osiva. Dále byly rozebrány možné způsoby detekce patogenů v semenech a rostlinách. Byl objasněn princip polymerázové řetězové reakce (PCR), její využití, možné obměny a optimalizace metod. Důraz byl kladen na metodu kvantifikace pomocí real-time PCR a determinace druhů pomocí PCR-RFLP. Součástí literární rešerše byla také charakteristika patogenů rodu Tilletia, jejich biologické vlastnosti a ekologické nároky. V praktické části byl vyhodnocen test klíčivosti se semeny pšenice seté (Triticum aestivum, L.) a pšenice špaldy (Triticum spelta, L.) ošetřenými pomocí technologie E-VENTUS. Klíčivost takto ošetřených semen nebyla snížena oproti neošetřeným variantám, semena pšenice seté ozimé ošetřené metodou E-VENTUS dokonce klíčila zpočátku rychleji než v neošetřené variantě. Dále byla provedena determinace druhů rodu Tilletia pomocí PCR. Byly identifikovány vzorky obsahující patogeny druhů Tilletia caries a Tilletia controversa. Byl založen biologický test s osivem různých variant ošetření včetně negativní kontroly. Infekce klíčních rostlin však nebyla úspěšná. Proto byly v rámci přípravy metodiky kvantifikace patogenů pouze optimalizovány podmínky reakce na vzorcích kalibrační křivky, které vycházely z DNA izolované ze snětivých hálek přítomných v osivu.

Abstract:

This work deals with the effectiveness of pre-sowing seed treatment in relation with the occurrence of seed-borne pathogens, focusing on genus Tilletia. The theoretical part describes the issue of seed treatment. The conditions of the correct choice of the treatment agent, treatment quality and its testing were discussed, as well as the possibilities of physical methods of seed treatment. Furthermore, the available methods for detection of pathogens in seed and plants were discussed. The principle of polymerase chain reaction (PCR), its use, possible variations and optimizations of this method were clarified. The emphasis was placed on quantification by real-time PCR and species determination by PCR-RFLP. The literature review also included the characteristics of pathogens of genus Tilletia, their biological properties and ecological demands. In the practical part, the germination test was performed on seed samples of common wheat (Triticum aestivum, L.) and spelt wheat (Triticum spelta, L.) treated by the E-VENTUS method. The germination of the treated seeds was not reduced in comparison with untreated variant. Furthermore, the determination of Tilletia species by PCR was performed. Samples containing pathogens of Tilletia caries and Tilletia controversa were identified. A biological test was established with seeds of various treatment variants, including a negative control. However, the infection of seedlings was not successful. Therefore, in the preparation of the methodology for the quantification of pathogens, the reaction conditions were optimized on the samples of the calibration curve, which were based on DNA isolated from the bunt balls present in infected seed lot.
