Effect of diclofenac on germination and growth of seedlings

Abstract:

Životní prostředí je stále intenzivněji kontaminováno pseudoperzistentními polutanty. Novou, významnou skupinu tvoří léčiva, jejichž spotřeba meziročně stoupá, a tím se také zvyšuje jejich vstup do prostředí. Mezi jedno z nejčastěji používaných nesteroidních protizánětlivých léčiv, které je ve velké míře detekováno v prostředí vzhledem k jeho nedostatečné degradaci v čistírnách odpadních vod, patří diklofenak. Cílem diplomové práce bylo posoudit vliv zvyšující se koncentrace diklofenaku (0,1; 10; 100; 1000 µg/l) na klíčivost semen vybraných druhů kulturních plodin (kukuřice setá - Zea mays L., cibule kuchyňská - Allium cepa L., hořčice bílá - Sinapis alba L., rajče keříčkovité - Solanum lycopersicum L., salát setý - Lactuca sativa L. a hrách setý - Pisum sativum L.) a zhodnotit jeho vliv (0,1; 1; 5; 10 mg/l) na růst (délka primárního kořene a stonku, listová plocha a hmotnost sušiny), biochemické (obsah chlorofylu a, b a karotenoidů a jejich poměr) a fyziologické parametry (fluoresecnce chlorofylu: FV/FM, ΦII, NPQ) hrachu setého. Přítomnost diklofenaku v kultivačním roztoku neovlivnila klíčivost semen s výjimkou cibule kuchyňské, u které bylo významné snížení klíčivosti semen zaznamenáno při koncentraci (5 a 10 mg/l). Hodnocení vlivu diklofenaku na rostliny hrachu setého proběhlo po 20 dnech kultivace. Významné snížení růstových parametrů bylo zaznamenáno až při vyšším stupni zatížení (5 a 10 mg/l). Obsah fotosyntetických pigmentů ani parametry fluorescence chlorofylu nebyly významně ovlivněny. Z výsledků je patrné, že vyšší zatížení prostředí diklofenakem může negativně ovlivnit růst rostlin. Je potřeba léčiva testovat na více úrovních, jelikož standardizované testy mají nižší citlivost.

Abstract:

The environment is getting more intensively contaminated with pseudopersistant pollutants. A new and important class of pollutants are pharmaceuticals, the consumption of which increases every year and leads to an increased input into the environment. One of the most intensively used nonsteroidial antiinflammatory drug, diclofenac, is often found in the environment due to its poor elimination during waste water treatment processes. The aim of this thesis was to assess the effect of increasing concentrations of diclofenac (0,1; 10; 100; 1000 µg/l) on the germination of seeds of selected crops (maize - Zea mays L., onion - Allium cepa L., white mustard - Sinapis alba L., tomato - Solanum lycopersicum L., lettuce - Lactuca sativa L. and pea - Pisum sativum L.) and to assess its effect (0,1; 1; 5; 10 mg/l) on growth (the length of primary root and shoot, leaf area and dry mass), biochemical parameters (content of chlorophyll a, b and carotenoids) and physiological parameters (fluorescence of chlorophyll: FV/FM, ΦII, NPQ) of pea plants. The presence of diclofenac in the culture solution didn’t significantly influence the germination of seeds with the exception of onion, where a decrease in germination rate was found with the concentration 5 and 10 mg/l. The effect of diclofenac on pea plants was evaluated after 20 days of cultivation. It has been shown to significantly decrease the growth parameters by increased load of diclofenac (5 and 10 mg/l). Neither the content of photosyntetical pigments nor the parameters of chlorophyll fluorescent were affected. The results show that the increased load of diclofenac in the environment may negatively affect the growth of plants. There is a need for the drugs to be tested on multiple levels, since standardized tests have lower sensitivity.
