Use of bacterial colorimetric test for an evaluation of surface waters acute toxicity

Abstract:

Bakalářská práce se zabývá hodnocením akutní toxicity povrchových vod pomocí bakteriálního kolorimetrického testu. Pro test byl použit bakteriální kmen Rhizobium meliloti, který ve svém dýchacím řetězci redukuje barvivo MTT. Redukce je doprovázena barevnou změnou, která závisí na množství přítomných toxických látek. Teoretická část obsahuje stručnou specifikaci problematiky znečištění povrchových vod a dosažených pokroků v oblasti biotestů k detekci polutantů. Je zde dále uvedena i definice a vymezení pojmů oboru ekotoxikologie. Praktická část je zaměřená na hodnocení akutní toxicity standardních látek a vzorku povrchové vody řeky Olše pomocí bakteriálního kolorimetrického testu. Experimentální data byla vyhodnocena použitím lineární nebo logaritmické regrese. Byla stanovena hodnota IC50, která uvádí koncentraci toxické látky způsobující 50% inhibici redukce.

Abstract:

The bachelor thesis is based on an evaluation of surface waters acute toxicity using a bacterial colorimetric test. A bacterium phyla Rhizobium meliloti, which can reduce the MTT dye in its respiratory chain, was used for this test. This reduction is accompanied by a colour change dependent on the amount of occurred toxic chemicals. The theoretic part consists of a short specification of the surface water pollution issue and the achieved progress in bioassays for pollutants detection. There are also noted the definitions of the field of ecotoxicologia. The practical part is focused on an evaluation of acute toxicity of standard solutions and a surface water sample of river Olše with a bacterial colorimetric test. Experimental data were evaluated using linear or logarithmic regression. The IC50 value, which shows a 50% inhibition concentration reduction, was established.
