The use of MALDI-MS in the taxonomy of lactic acid bacteria

Abstract:

První část diplomové práce je tvořena literární rešerší na téma využití MALDI hmotnostní spektrometrie v mikrobiologii a věnuje se taxonomickému vývoji v rámci rodu Lactobacillus. MALDI MS se využívá již několik desítek let k analýze různých látek. Své místo nalezla i v mikrobiologii, jako rychlá a spolehlivá metoda jak v klasifikaci, tak i identifikaci bakterií, archeí, virů a hub. Laktobacily jsou gram-pozitivní bakterie vyskytující se v mnoha prostředích. Člověk je využívá ve výrobě a konzervaci potravin a také jako zdraví prospěšné organizmy – probiotika. Taxonomie v rámci tohoto rodu je proměnlivá, neustále jsou popisovány nové druhy a staré reklasifikovány. Klasické fenotypové metody pro jejich rozlišení mnohdy nestačí. Experimentální část práce si klade za cíl zhodnotit možnosti uplatění metody MALDI-MS v taxonomii těchto bakterií a posoudit vliv kultivačních podmínek (médium, teplota a délka kultivace) na výsledná hmotnostní spektra a zařazení jednotlivých druhů resp. kmenů v dendrogramu.

Abstract:

The first part of this diploma thesis comprise literary search about MALDI mass spectrometry usage in microbiology and it is focused on taxonomic development in the genus Lactobacillus. The mass spectrometry has been used for analysis of various substances for many years. It has found its place also in microbiolgy as fast and reliable method in classification and identification of bacteria, archaea, viruses and funghi as well. Lactobacilli are gram-positive bacteria living in many environments. Man uses them in food industry and also as beneficial organisms – probiotics. Taxonomy in this genus is very changable, new species are described and old ones are being reclassified permanently. Classic fenotypic methods are often not enough for their differentiation. The goal of the experimental part of this thesis is to evaluate the posibilities of using MALDI-MS in taxonomy of these bacteria and to asses an influence of cultivation conditions (the medium, the temperature and the lenght of cultivation) on final mass spectra and placement of every single species or strain in the dendrogram.
