Molecular-biological methods in archaeology

Abstract:

Tato habilitační práce shrnuje výsledky na poli mé práce s historickými proteiny a DNA. Zabýval jsem se identifikací kostí, zbytků potravin na keramice a také mitochondriální DNA (mtDNA) prasat. Imunologická metoda identifikace proteinů se na historické proteiny zkoušela v minulosti s nevyhovujícími výsledky, protože se neuvažovala degradace proteinové struktury během času. Pokud se použily protilátky na denaturovanou formu proteinů, bylo možné tuto metodu využít velmi spolehlivě. Podobně v minulosti prováděné izolace proteinů z porézní keramiky vedly k poškození proteinů, ale podařilo se vypracovat velmi jednoduchou metodu k získání těchto proteinů a jejich následným analýzám. Byla taqké vypracována metodika na odlišení proteinů z trusu bukální sliznice prasat (Sus scrofa), a vařených historických proteinů, a tak odlišit kontaminace a falešně pozitivní výsledky. Trus se dostává do keramiky v zemi zřejmě pomocí vody a může pocházet z historické doby, nebo ze současného zemědělství. Podařila se prokázat řada nových faktů, či některé potvrdit. Výsledky např. potvrzují hypotézu, že keramika se ve starších dobách používala opakovaně, zatímco ve středověku byla levná porézní keramika jednorázová záležitost jako dnes např. plasty. Ve středověkých městech byla koza zřejmě běžné domácí zvíře a převažovala konzumace kozího mléka, zatímco venkovské oblasti používaly kravské i kozí. Hodně mých výsledků se týkalo doby bronzové, kde se např. podařilo prokázat úpravy masa na keramických plotnách, nebo výrazné zastoupení mléka v potravě, přestože konzumenti by neměli být schopni trávit mléčný cukr laktózu. Mnohdy překvapuje nepřítomnost obilí v keramice, kde se předpokládaly obilné kaše. Ze sekvenování historické mtDNA je možno vyvodit, že na území České republiky byla zřejmě od neolitu do novověku jen jedna linie prasat (Sus scrofa), zřejmě domestikovaná z místních prasat divokých, protože tato linie stále mezi evropskými divokými prasaty existuje. Studium minulosti pomocí molekulárně biologických metod je s nezbytnou opatrností použitelné pro rekonstrukce stravy a v širším měřítku pro rekonstrukce antropogenně ovlivněných životních prostředí v minulosti.

Abstract:

This habilitation thesis summarizes results of my work with ancient proteins and DNA. I worked on the identification of bones, food residues on ceramics and mitochondrial DNA (mtDNA) of pigs. The immunological method of protein identification has been tried on ancient proteins in the past with unsatisfactory results because the degradation of the protein structure over time was not considered. When antibodies to a denatured form of proteins were used, this method could be used very reliably. Similarly, the isolation of proteins from porous ceramics in the past led to protein damage, but it was possible to develop a very simple new method to obtain these proteins and their subsequent analyses. A methodology was also developed to distinguish proteins from the faeces of the buccal mucosa of pigs (Sus scrofa) and boiled historical proteins, and thus distinguish contamination and false positive results. The dung gets into the pottery in the country, apparently with the help of water, and can come from historical times or from contemporary agriculture. It was possible to prove a number of new facts, or to confirm some. The results, for example, confirm the hypothesis that ceramics were used repeatedly in older times, while in the Middle Ages cheap porous ceramics were a one-time thing like today, for example, plastics. In medieval cities, the goat was apparently a common domestic animal and the consumption of goat's milk prevailed, while rural areas used both cow's and goat's milk. Many of my results related to the Bronze Age. In this period, for example, it was possible to demonstrate the preparation of meat on ceramic plates, or the significant presence of milk in food, even though consumers should not be able to digest the milk sugar lactose. The absence of grains in ceramics, where grain porridges were assumed, is often surprising. From the sequencing of historical mtDNA, it is possible to conclude that there was probably only one line of domestic pigs (Sus scrofa) in the territory of the Czech Republic from the Neolithic to the modern era, apparently domesticated from local wild pigs, because this line still exists among European wild pigs. The study of the past using molecular biological methods is, with the necessary caution, applicable to diet reconstructions and, on a wider scale, to reconstructions of anthropogenically influenced environments in the past.
