Functional analysis of temperature-sensitive mutants of tumor suppressor p53

Abstract:

Protein p53 je nádorový supresor, který v odpovědi na buněčný stres vyvolává zástavu buněčného cyklu nebo programovanou buněčnou smrt. To vede k eliminaci premaligních a maligních buněk a k prevenci vzniku nádorů. Ztráta funkce proteinu p53 hraje významnou roli v procesu karcinogeneze u většiny typů nádorů. Více než 50% nádorů obsahuje nefunkční protein p53 v důsledku bodové mutace v genu p53. Reaktivace nádorového supresoru p53 se v poslední době jeví jako jedna z možných strategií specificky cílené, protinádorové terapie. Pomocí funkční metody FASAY („functional analysis of separated alleles in yeast“), která se v naší laboratoři využívá k rutinní analýze statutu proteinu p53 u různých typů nádorů, byl sestaven soubor 23 teplotně závislých mutantů p53. Za využití kvasinkového expresního systému jsme podrobně analyzovali funkční a strukturní vlastnosti mutantů v našem souboru. Studovali jsme transaktivační schopnosti jednotlivých mutantů ve vztahu k teplotě kultivace a k sekvenci různých responzivních elementů. Jednotliví mutanti se mezi sebou lišili mírou teplotní závislosti, tj. teplota, při které došlo k úplné nebo částečné obnově jejich transaktivačních funkce, byla různá. Většina teplotně závislých mutantů vykazovala tzv. diskriminativní charakter, tj. lišila se mírou své aktivity vůči různým responzivním elementům. Při studiu strukturních vlastností teplotně závislých mutantů s využitím konformačně specifických protilátek jsme nenašli žádnou souvislost mezi mírou transaktivace a konformačními vlastnostmi jednotlivých mutantů p53. U 13 mutantů p53 jsme pozorovali částečnou reaktivaci jejich transaktivačních schopností po působení chemické látky amifostinu. Pomocí počítačového molekulového modelování jsme zjistili, že většina mutantů p53 (10/13), kteří jsou reaktivováni po působení amifostinu, obsahuje mutaci v oblasti genu p53, která kóduje hydrofobní β sendvičovou strukturu DNA vazebné domény proteinu. Předpokládáme, že amifostin tuto strukturu stabilizuje, čímž je částečně nebo zcela obnovena funkce celé domény. Funkčními a strukturními analýzami jsme prokázali, že teplotně závislí mutanti proteinu p53 tvoří značně heterogenní skupinu proteinů. Mezi jednotlivými mutanty existuje značná variabilita v konformačních a funkčních vlastnostech a ve schopnostech být za určitých podmínek reaktivováni. Výsledky naší práce potvrzují, že protein p53 je konformačně flexibilní a jeho funkci lze reaktivovat pomocí malých molekul jako je amifostin. Podrobná znalost funkčních a strukturních vlastností jednotlivých mutantů p53 a jejich stratifikace může být užitečná pro lepší pochopení účinků specifických mutantů na procesy vedoucí k transformaci nádorové buňky, při predikci průběhu nádorového onemocnění, jeho léčbě a také při vývoji látek schopných obnovy standardní funkce proteinu p53.

Abstract:

The p53 protein is a tumor suppressor which in response to cell stress promotes cell cycle arrest or programmed cell death. This leads to elimination of pre-malignant and malignant cells and to prevention of tumor formation. Loss of the p53 protein function plays an important role in carcinogenesis of the majority of tumor types. More then 50% of human tumors contains non-functional p53 protein due to mutation in the p53 gene. Reactivation of p53 is promising therapeutical approach to specifically combat cancer. Using functional method FASAY („functional analysis of separated alleles in yeast“), which is used in our laboratory for routine analysis of p53 status from different type of tumors, the collection of 23 temperature dependent p53 mutants was set up. Using the yeast expression system, we analyzed the functional and structural features of our p53 mutants in detail. We studied dependency of transactivation abilities of individual mutants on cultivation temperature and responsive elements specificity. We found various temperature dependent mutants differing in their temperature dependency, transactivation abilities of these mutants were partially or fully restored in different temperatures. Majority of temperature dependent mutants also differed in the level of their transactivation ability toward various types of responsive elements derived from the promotor regions of genes bax, p21 and the RGC documenting their discriminative character. We found no correlation between the transactivation rate and conformation of the p53 temperature-dependent mutants using conformation specific antibodies. In 13 of 23 temperature sensitive p53 mutants, we observed partial transactivation reactivation upon treatment with amifostine. The most of the p53 mutations (10/13) reactivated by amifostine were located in the part of the p53 gene coding for hydrophobic β-sandwich structure of the DNA-binding domain. We suppose that amifostine stabilizes this structure and thereby the function of the whole domain is partially or fully restored. By functional and structural analysis we documented, that the temperature-dependent mutants are considerably heterogeneous group of proteins. There is significant variability in conformational and functional features of the mutants and in their reactivation ability. Results of our work confirm that the p53 protein is conformationally flexible and its function is possible to restore by small molecules such as amifostine. Detailed knowledge of functional and structural abilities of individual p53 mutants and their stratification could be useful for better understanding of the biological effects of the mutants in processes of carcinogenesis, in prediction of the disease outcome, its treatment and in a development of the drugs able to restore the standard function of the protein.
