Study of ecortoxicological properties using screening genotoxicity bioassays

Abstract:

V současné době si velmi dobře uvědomujeme, že se chemické mutageny vyskytují v různých environmentálních matricích jako jsou pitná voda, povrchová voda, sedimenty, půdy, volné nebo vnitřní ovzduší. Zřejmá potřeba kvalitních monitorujících systémů či potřeba se chránit před vystavením mutagenních nebo karcinogenních látek vedla k vývoji celé řady biotestů za účelem detekce genotoxicity. Tato práce je zaměřena na studium ekotoxikologických vlastností s využitím testů genotoxicity a je zde popsána široká škála odlišných metodických postupů detekce genotoxicity. Pro studium vlastností jak čistých chemických látek, tak environmentálních směsí bylo využito dvou velmi známých bakteriálních testů (umuC test a SOS Chromotest) a také nový systém využívající eukaryotické buňky kvasinek. Toxafen je směs několika stovek látek. Vzhledem k jeho vysoké persistenci a bioakumulaci se jeho toxické vlastnosti velmi intenzivně studují. V obou použitých testech byly pro genotoxickou aktivitu pozorovány závislosti dávka-účinek. SOS Chromotest dokonce potvrdil statistickou významnost genotoxického potenciálu této látky. Kromě technického toxafenu jsme studovali také vlastnosti UV ozařovaného toxafenu. Vzhledem ke snižující se koncentraci 22 hlavních kongenerů toxafenu v ozařované směsi a současného zvýšení genotoxické odpovědi můžeme předpokládat vznik mnohem nebezpečnějších produktů degradace ve srovnání s původní směsí technického toxafenu. Pro detekci genotoxických vlastností byl v současné době vyvinut zcela nový systém, který využívá kmen Saccharomyces cerevisiae s GFP proteinem. Tato práce potvrzuje schopnost využít tento GFP test pro studium toxických i genotoxických vlastností environmentálních polutantů. Testováno bylo 20 azaarenů, přičemž výsledky naznačují, že azaareny mají vyšší toxickou i genotoxickou aktivitu než jejich homocyklické analogy. Také bylo provedeno srovnání testu s bakteriálními biotesty. Toto srovnání ve většině případech ukazuje na srovnatelnou přesnost co do pozitivních a negativních odpovědí, nicméně poukazuje na mnohem vyšší citlivost co do dosažených efektivních koncentrací. Dále byla studována schopnost využití pasivní vzorkovací metody v procesu hodnocení přímé genotoxicity vzorků volného ovzduší. 11 z celkových 20 vzorků z brněnských lokalit vykázalo pozitivní odpověď v testu genotoxicity – SOS Chromotestu. Potvrdili jsme velmi dobré využití metody pasivního vzorkování jak pro sledování časoprostorových změn úrovně kontaminace prostředí, tak velmi užitečnou funkčnost metody při hodnocení genotoxického potenciálu. Závěrem jsme studovali aplikaci rozličných přístupů v procesu hodnocení genotoxicity reálných environmentálních vzorků. S využitím SOS Chromotestu bylo demonstrováno, že výběr dokonalejších statistických modelů může v procesu hodnocení zdravotních rizik představovat mnohem účelnější postup odhadu negativních efektů vedoucích z expozice volným ovzduším.

Abstract:

Today, we are well aware of the fact that chemical mutagens are present in a variety of complex environmental matrices, including drinking water, surface water, aquatic sediment, soil, indoor air, and ambient urban air. The obvious need for sensitive monitoring of environmental pollutants, to protect against mutagenic or carcinogenic effects of these substances, has led to the development of numerous bioassays for genotoxicity detection. This work is focused on study of ecotoxicological properties using screening genotoxicity bioassays. A variety of different methods for genotoxicity detection has been reported. We selected two well known bacterial tests (umuC test and SOS Chromotest) and new eukaryotic yeast assay for study both pure chemicals and also environmental complex mixtures. Toxaphene is a complex mixture of several hundred components. Because of its high persistence and bioaccumulative potency its toxic effects are at great interest. Both in the SOS Chromotest and the umuC test there was observed dose-response relationship of genotoxic activity. Using SOS Chromotest toxaphene was clearly marked as significant genotoxin. Besides technical toxaphene, we also studied the genotoxic properties of UV-irradiated toxaphene. Because of decreasing concentration of detectable 22 congeners and simultaneously increasing genotoxic potency of irradiated water samples, we suppose formation of toxic toxaphene products that may have high genotoxic properties compared to technical toxaphene. Saccharomyces cerevisiae tester strains have been developed for detection of genotoxic potential with yeast green fluorescent protein (GFP). We presented evidence that the GFP assay can be successfully used for studying toxicity and genotoxicity of environmental pollutants. We established both the toxic and genotoxic potential of 20 azaarenes and results indicate azaarenes have higher toxic and genotoxic potencies than their homocyclic analogues. We also compared GFP assay with bacterial toxicity and genotoxicity bioassay and this comparison indicates that in most cases the GFP assay is apparently of comparable sensitivity to the bacterial toxicity or genotoxicity tests, but much more sensitive with respect to the effective concentration values. We also investigated the capability of passive air sampling technique to be employed in the evaluation of direct genotoxicity of ambient air samples. The SOS Chromotest revealed direct mutagenicity answers in 11 from 20 samples from Brno localities. We reported not only a very good capability of the passive air samplers to reflect the spatial fluctuation in contamination levels in the ambient air but the feasibility of using this method for the direct genotoxic potential assessment. Finally, we were focused on application of different evaluative approaches for genotoxicity assay on real environmental samples. For genotoxicity screening in vitro SOS Chromotest was used. We demonstrated that choosing more sophisticated statistical models suggest the usefulness of the methodological approach for prediction potential hazards related to exposure of urban air within health risk assessment.
