Role of plant telomere proteins in cell regulations

Abstract:

Telomery jsou dynamické nukleoproteinové struktury, které chrání konce lineárních eukaryotických chromozomů. Nechráněná telomerická DNA by mohla být rozpoznána například jako dvouřetězcový DNA zlom, a proto jednou z možností, jak předcházet rozpoznání telomerické DNA reparačními mechanismy, je vazba specifických proteinů. Zatímco interaktom ochranného komplexu proteinů u savců byl dobře popsán, informace o funkci a vzájemných interakcích rostlinného komplexu chránícího telomery byly odhaleny teprve nedávno. Proteiny asociované s telomerami ovlivňují nejen aktivitu a funkci telomer, ale také regulují jejich vazbu s enzymem – telomerázou, který se podílí na udržování konců chromozomů. Telomere Repeat Binding (AtTRB) proteiny jsou proteiny specifické pro rostliny a naše vědecká skupina zjistila, že tyto proteiny rostliny Arabidopsis thaliana váží telomerické repetice díky jejich Myb doméně typu telobox in vitro. Rostlinné AtTRB jsou homology savčích TRF (Telomere Repeat Factor) proteinů, jež jsou základními komponenty shelterin komplexu podílejícího se na ochraně telomer i u člověka. V rámci svého studia jsem se podílela na několika původních vědeckých pracích, ve kterých naše skupina prokázala, že AtTRB proteiny jsou kolokalizovány s rostlinnými telomerami in situ a in vivo, a fyzicky interagují s katalytickou podjednotkou rostlinné telomerázy (AtTERT). Dále jsme prokázali, že AtTRB1 protein je in vivo také asociován s promotory genů uplatňujících se při biogenezi ribozomů. Charakterizovali jsme celou řadu proteinů purifikovaných s AtTERT a mezi nimi jsme identifikovali také homology RuvBL1 (Pontin) a RuvBL2 (Reptin) proteinů, které u savců ovlivňují biogenezi telomerázy. V následující práci se nám podařilo charakterizovat jejich rostlinné homology nazývající se AtRuvBL1 a AtRuvBL2a a odhalili jsme, že AtRuvBL1 protein asociuje in vivo v jadérku s rostlinnou katalytickou podjednotkou telomerázy (AtTERT). Na rozdíl od situace u savců, interakce mezi AtRuvBL a AtTERT je u rostlin nepřímá a zprostředkovávají ji AtTRB proteiny. Navíc se ukázalo, že skládání telomerázy je u rostlin dynamický proces, stejně jako u savců. Rostlinný homolog savčího proteinu dyskerinu, nazývající se AtCBF5, nepřímo interaguje s AtTRB proteiny, ale ne s AtRuvBL proteiny a také fyzicky interaguje s AtPOT1a proteinem – rostlinným homologem savčího POT1. Rostliny heterozygotní v genech AtRuvBL se vyznačují sníženou aktivitou telomerázy, což také poukazuje na jejich zapojení do skládání telomerázy u rostlin. Naše studie zaměřená na charakterizaci rostlinného nucleolinu pomohla odhalit roli jadérka, kde není lokalizována pouze mašinérie podílející se na skládání telomerázy, ale také mechanismy účastníci se prostorové organizace chromozomů a udržování telomer. Tato práce poskytuje přehled dosud zjištěných faktů v oblasti proteinů asociovaných s rostlinnými telomerami a telomerázou.

Abstract:

Telomeres are dynamic nucleoprotein structures and protect linear eukaryotic chromosomal ends. Unprotected telomeric DNA could be recognized as double strand breaks. One way how to prevent recognition of telomeres by DNA repair machinery is the recruitment of specific capping proteins. While the interactome of mammalian telomeric protein complex has been well described, data concerning the functions and interactions of the plant telomere-associated proteins arised only recently. Telomere-associated proteins play important role not only in regulation of the function and activity of the plant telomeres, but they also regulate access of plant telomerase enzyme. Telomere Repeat Binding (TRB) proteins are plant specific proteins. During the past decade, we have shown that Arabidosis thaliana TRB proteins specifically bind telomeric repeats through their Myb domain of the telobox type in vitro. Plant TRB proteins are homologues of TRF (Telomere Repeat Factor) proteins, which are members of mammalian shelterin protein complex. Shelterin protein complex safeguards mammalian telomeric ends. As part of my studies, I have participated in several scientific publications in which our group has demonstrated that AtTRB proteins are colocalized with plant telomeres in situ and in vivo and physically interact with catalytic subunit of plant telomerase called AtTERT. We have further revealed the in vivo association of AtTRB1 protein with promoter regions of genes involved in ribosome biogenesis. We have characterized a variety of proteins co-purificated with AtTERT and identified RuvBL1 and RuvBL2 proteins, which play role in mammalian telomerase biogenesis. In the subsequent work, we described plant homologues of RuvBL proteins called AtRuvBL1 and AtRuvBL2a and revealed association between AtRuvBL1 and AtTERT in vivo. In contrast with mammals, AtRuvBL1-AtTERT interaction is indirect and it is rather mediated by one of the AtTRB proteins. Next, we have shown that telomerase assembly is dynamic process in plants as well as in mammals. Plant homologue of mammalian dyskerin protein, called AtCBF5, indirectly associates with all three AtTRB proteins, but not with AtRuvBL proteins and physically interacts with AtPOT1a – plant homologue of mammalian shelterin POT1. Plants heterozygous in AtRuvBL genes showed reduced telomerase activity indicating their potential involvement in telomerase assembly in A. thaliana. Our study focused on characterization of plant nucleolin protein helped to reveal that plant nucleolus is not only place of telomerase assembly, but also contains mechanisms for spatial organization of chromosomes and telomere maintenance. This thesis provides overview of so far found facts in the field of associated proteins connected with plant telomeres and telomerase.
