The role of tumor necrosis factor-inducible gene 6 protein in the regulation of the specific immune response

Abstract:

Protein genu 6 indukovatelný tumor nekrotizujícím faktorem (TSG-6) je multifunkční protein, který je sekretován zejména v zánětlivých ložiscích, kde hraje zásadní roli v regulaci imunitního systému. Dosavadní studie jej spojují s protizánětlivým účinkem, kterého dosahuje modulací imunitní odpovědi. Zda však TSG-6 ovlivňuje adaptivní imunitní systém, a případně jakým mechanismem toho dosahuje, nebylo zatím předmětem téměř žádného výzkumu. Z tohoto důvodu se teoretická část práce zaměřuje na popsané efekty TSG 6 na imunitní odpověď s důrazem na specifickou imunitu. V experimentální části si klade za cíl identifikovat vliv TSG-6 na buňky specifické imunity, T pomocné (Th) lymfocyty, izolované z periferní krve zdravých dárců. Po ovlivnění buněk proteinem TSG-6 byla měřena jejich viabilita, proliferace, povrchová exprese aktivačních molekul a produkce prozánětlivého cytokinu interferonu gamma (IFN-γ). Výsledky ukázaly, že určité koncentrace TSG-6 potlačily aktivaci Th lymfocytů, a to přes modulaci jejich fenotypu. Z proběhlých experimentů lze shrnout, že TSG-6 ovlivňuje buňky specifické imunity, což je důležitý a nový poznatek ve výzkumu tohoto proteinu. Zároveň získané výsledky přispívají k možnostem terapeutické aplikace TSG-6 u onemocnění spojených s patologií Th lymfocytů, jako je například revmatoidní artritida, diabetes I. typu či ulcerózní kolitida.

Abstract:

Tumor necrosis factor-stimulated gene 6 (TSG-6) is a multifunctional protein that is secreted mainly at sites of inflammation where it has been found to play a crucial role in immune regulation. Recent research has suggested that TSG-6 plays an anti-inflammatory role there by modulating the immune response. However, whether TSG-6 regulates the adaptive immune system, and if so by which mechanism, has not been investigated in almost any way. Therefore, the theoretical part of this work is focused on the described effects of TSG-6 on the immune response with a focus on specific immunity. In the experimental part, the objective was to discover effects of TSG-6 on the class of specific immunity, activated T helper (Th) cells, isolated from the blood of healthy donors. After treatment with TSG 6, the viability, proliferation, activation markers, and production of pro inflammatory cytokine interferon gamma (IFN-γ) was measured. The results showed that certain concentrations of TSG-6 suppressed the activation of Th lymphocytes through the modulation of their phenotype.The experiments conducted suggest that TSG-6 has an impact on cells of specific immunity, which is an important and new finding in the research on this protein. Furthermore, these results contribute to the potential therapeutic application of TSG-6 in diseases associated with Th lymphocyte pathology, such as rheumatoid arthritis, type 1 diabetes, and ulcerative colitis.
