Abstract:

Plicní hypertenze je závažné kardiovaskulární onemocnění, zvyšující mortalitu pacientů. Plicní hypertenze může být spojována mimo jiné i s aktivitou galektinu-3. Galektin-3 je schopen podporovat proliferaci hladkých svalových buněk mediální vrstvy cévní stěny a tím zvyšovat odpor cév vůči proudící krvi. Plicní hypertenze může být vyvolána vystavením organismu hypoxii, a proto byl zkoumán rozdíl mezi buňkami izolovanými z plicních arterií potkanů vystavených hypoxickým nebo normoxickým podmínkám. Byla charakterizována adheze a růst normoxické primokultury a zjištěna čistota normoxických a hypoxických primokultur. Dále byly porovnány normoxické a hypoxické primokultury v závislosti na expresi typických hladkosvalových markerů, galektinu 3 a kolagenu I. Zároveň byla také porovnána exprese těchto markerů v plicní arterii, pravé srdeční komoře a plicích potkanů vystavených hypoxii nebo normoxii. Galektin-3 přispívá ke tvorbě plicní hypertenze, a proto byla rovněž testována cytotoxicita inhibitorů galektinu-3 v normoxických i hypoxických primokulturách.

Abstract:

Pulmonary hypertension is a serious cardiovascular disease, increasing patient mortality. Pulmonary hypertension can be associated, among other things, with galectin-3 activity. Galectin-3 is able to support the proliferation of smooth muscle cells of the medial layer of the vascular wall and thus increase the resistance of the vessels to the flowing blood. Pulmonary hypertension can be induced by exposing the organism to hypoxia, so the difference between cells isolated from pulmonary arteries of rats exposed to hypoxic or normoxic conditions was investigated. Adhesion and growth of normoxic primocultures were characterized and the purity of normoxic and hypoxic primocultures was determined. Furthermore, normoxic and hypoxic primocultures were compared with respect to the expression of typical smooth muscle markers, galectin-3 and collagen I. At the same time, the expression of these markers in the pulmonary artery, right ventricle and lungs of rats exposed to hypoxia or normoxia was also compared. Galectin-3 contributes to the formation of pulmonary hypertension, and therefore the cytotoxicity of galectin-3 inhibitors was also tested in normoxic and hypoxic primary cultures.
