Bioinformatic analysis of proteins interacting with DNA triplexes

Abstract:

Tato práce se zabývá triplex-vazebnými proteiny. Teoretická část je zaměřena na popis struktury a funkci jednotlivých triplex-vazebných proteinů. Dále obsahuje také charakteristiku, rozdělení a biologickou roli triplexových struktur. Praktická část se zabývá bioinformatickou analýzou aminokyselinového složení devatenácti triplex-vazebných proteinů. Na základě těchto analýz bylo zjištěno, že tyto proteiny, stejně jako proteiny vázající jiné nekanonické struktury, obsahují ve vysokém procentuálním zastoupení kyselinu glutamovou (E), leucin (L) a serin (S) a v nízkém zastoupení cystein (C), histidin (H), methionin (M), tryptofan (W) a tyrosin (Y). Proteiny vázající triplexy, G-kvadruplexy, Z-DNA nebo křížové struktury jsou obohaceny o kyselinu asparagovou (D) a ochuzeny o cystein (C). Dále byl odvozen společný sekvenční motiv 19 triplex-vazebných proteinů (HEEEERELQEEQLEQQEVQ).

Abstract:

This work deals with triplex-binding proteins. The theoretical part is focused on the structure and function of individual triplex-binding proteins. It also contains characteristics, classification, and biological role of triplex structures. The experimental part is focused on bioinformatic analyses of the amino acid composition of 19 triplex-binding proteins. Based on these analyzes, it was found that triplex-binding proteins, as well as proteins that bind to other non-canonical structures, are predominantly composed of glutamic acid (E), leucine (L), and serine (S) and are abundant in cysteine (C), histidine (H), methionine (M), tryptophan (W) and tyrosine (Y). Proteins that bind to triplexes, G-quadruplexes, Z-DNA, or cruciforms are enriched in aspartic acid (D) and depleted of cysteine (C). Furthermore, a common sequence motif of 19 triplex-binding proteins (HEEEERELQEEQLEQQEVQ) was discovered.
