Genetic analysis of putative sensor histidine kinases from Brassica napus

Abstract:

Cytokininy (CK) jsou fytohormony působící na mnohé vývojové a adaptační procesy rostlin, především pak buněčné dělení a diferenciaci. Jejich metabolizmus sehrává důležitou úlohu při regulaci růstu a vývoje kořene i nadzemní části rostliny. Molekuly tohoto regulátoru jsou vnímány prostřednictvím transmembránových proteinů – senzorových histidinkináz (HK). Ty byly poprvé identifikovány a důsledně popsány u modelového organizmu Arabidopsis thaliana v podobě tří homologů označovaných jako AHK2, 3 a 4. Cílené sekvenování rostlinných genomů moderními metodmi v posledních letech, stejně jako zavedení spolehlivých bioinformatických analytických nástrojů, umožnilo vyhledávání ortologů AHK i u hospodářsky významných plodin jako jsou kukuřice (Zea mays) nebo rýže (Oryza sativa). Genom brukve řepky – Brassica napus, není zatím veřejně dostupný prostřednictvím databází. Na základě výrazné homologie s A.thaliana se podařilo hybridizací s genomovou knihovnou B. napus identifikovat klony bakteriálních umělých chromozómů (BAC), nesoucích geny potenciálních receptorů CK (BnHK). V této práci popisuji metody reverzní genetiky a bioinformatické analýzy směřující k získání úplných sekvencí genomové DNA a cDNA všech predikovaných BnHK identifikovaných klonů BAC, dále metody klonování cDNA a transformace kvasinek touto cDNA za účelem potvrzení funkce identifikovaných genů in vivo.

Abstract:

Cytokinins are plant hormones which influence a lot of aspects of plant growth and development, mainly cell division and differentiation. Their metabolism plays a significant role in regulating the growth and development of the shoot and root system of plants. Cytikinins are perceived through the plant cell transmembrane proteins, histidine kinases (HK). For the first time, they were identified and thoroughly described in the model organism Arabidopsis thaliana and named AHK2, 3 and 4. Successful search for the AHK ortologues in significant crops such as Zea mays or Oryza sativa has been maid possible due to the application of modern automated sequencing techniques during the past few years, as well as due to the well-established and reliable tools of bioinformatics . The genome of another agriculturally attractive crop - Brassica napus, rapeseed –is not yet publicly available in electronic genome databases. Based on a considerable homology with A. thaliana, clones of bacterial artificial chromosomes (BAC), carrying genes of potential cytokinin receptors, BnHKs, have been identified in the genome library of B. napus. In this thesis I describe methods of reverse genetics and bioinformatics that will be used to obtain the full-length genomic DNA and cDNA.In addition, an established method to clone and express the recombinant cDNA in a heterologous yeast expression system is also described in order to prove fhe function of the identified genes in vivo.
