Monitoring of parameters monitoring the inflammation from the perspective of haematological laboratories

Abstract:

Problematika zánětu je v dnešní době velmi obsáhlé a diskutované téma. Zánětlivá reakce není způsobena pouze jediným procesem, ale kombinací mnoha faktorů působících na buňky, tkáně a celý organismus. Je to proces, který se snaží vyrovnat s vnějšími vlivy prostředí a ochránit organismus před nebezpečí infekce. V práci jsem se zaměřila pouze na malou část buněk, které jsou součástí obranného mechanismu organismu. V teoretické části práce se zabývám popisem průběhu zánětlivé reakce a buněčnými složkami, které jsou jeho součástí. Další částí práce je všeobecný souhrn automatických analyzátorů, metod, které jsou v dnešní době nejčastěji využívány a popisu hematologické linky Sysmex XN-9000 DI-60. Praktická část je věnována dvěma skupinám pacientů. Do první skupiny jsem zařadila dospělé pacienty. Do druhé skupiny jsem zařadila předčasně narozené děti. U pacientů v první skupině vyhodnocuji celkem čtyři parametry (leukocyty, nezralé granulocyty, granularitu neutrofilů a aktivované lymfocyty). U pacientů ve druhé skupině vyhodnocuji o jeden parametr navíc a to retikulované trombocyty. V konečné fázi práce jsem se věnovala porovnávání jednotlivých parametrů a jejich využitelnosti v klinické praxi. Jelikož byli pacienti pozorováni v čase, zvolila jsem retrospektivní sběr dat. Data jsem získala z laboratorního informačního systému (LIS). Vzorky byly měřeny v časovém intervalu od 2. 4. 2017 do 2. 3. 2018 pomocí hematologické linky Sysmex XN-9000 DI-60.

Abstract:

The issue of inflammation is a very extensive and discussed subject today. The inflammatory reaction is not caused by a single process but by a combination of many factors affecting cells, tissues and the whole organism. It is a process that strives to cope with the external influences of the environment and protect the organism from the danger of infection. In the thesis I focused only on a small part of the cells that are part of the body's defense mechanism. In the theoretical part of the thesis I deal with the description of the course of the inflammatory reaction and the cell components, which are part of it. Another part of the thesis is a general summary of automatic analyzers, methods that are currently used and described most commonly by the Sysmex XN-9000 DI-60. The practical part is devoted to two groups of patients. I have included adult patients in the first group. In the second group I included prematurely born children. The patients in the first group evaluate four parameters (leukocytes, immature granulocytes, granular neutrophils and reactive lymphocytes). In patients in the second group, one additional parameter and reticulated plasma cells are evaluated. At the final stage of my work I studied the comparison of individual parameters and their usability in clinical practice. As patients were observed over time I chose retrospective data collection. The data was obtained from the Information System Lab (LIS). The samples were measured in the time interval from 2. 4. 2017 to 2. 3. 2018 using the Sysmex XN-9000 DI-60 hematology line.
