Úloha fotosyntetických pigmentů při aklimaci asimilačního aparátu rostlin na různé mikroklimatické podmínky

The role of photosynthetic pigments in acclimation of the assimilation apparatus to different microclimatic conditions

Abstract:

Ve své disertační práci se zabývám vlivem vybraných abiotických faktorů, především tedy změnami v kvalitě dopadající radiace (a její intenzitě) a zvýšenou teplotou na vybrané druhy rostlin s důrazem na kvalitativní a kvantitativní zastoupení fotosynteticky aktivních pigmentů, mezi které (u vyšších rostlin) řadíme chlorofyly a karotenoidy (karoteny a xantofyly). Tyto pigmenty mají ve fotosyntetickém aparátu nenahraditelnou funkci a to nejenom v úloze absorpce dopadajícího světelného záření, ale také v ochraně fotosyntetického aparátu proti fotooxidativnímu poškození v podmínkách stresu. Mezi hlavní metodiky, kterým jsem se v rámci experimentální práce věnovala, patří absorpční spektrofotometrie a vysokoúčinná kapalinová chromatografie. V rámci identifikace derivátů chlorofylů byly používané metodiky okrajově rozšířeny o Ramanovu a hmotnostní spektrometrii a elektroforézu (SDS-PAGE, CN-PAGE). V rešeršní části práce je shrnut význam fotosyntetických pigmentů vyšších rostlin a jejich biosyntetické dráhy. V souvislosti s chlorofyly je rešerše zaměřena na detailní popis struktury a funkce enzymu Geranylgeranyl reduktázy, a také jeho souvislosti s funkčním stavem fotosyntetického aparátu a kumulací derivátů chlorofylů. Informace týkající se karotenoidů jsou pak zaměřeny na jejich význam v ochraně proti fotooxidativnímu poškození. Důraz je kladen na význam tepelné disipace nadbytečné excitační energie a ochrannou funkci xantofylového cyklu. V této části se také nacházejí informace shrnující vliv vybraných abiotických faktorů (spektrální kvality světla, UV záření, vysoké ozářenosti a teplotního stresu) na obsah těchto pigmentů u rostlin. Druhá část práce obsahuje motivaci vedoucí k uskutečnění experimentů a komentáře k přiloženým článkům, ve kterých jsme zabývali: 1) Identifikací derivátů chlorofylů kumulujících se u rostlin kultivovaných v podmínkách zeleného světla, 2) Zpětnou aklimací rostlin po jejich přenosu z podmínek vysokého světla, do podmínek po odeznění tohoto stresu, se zaměřením na podíl významu karotenoidů a UV-stínícíh látek v úloze ochrany fotosyntetického aparátu, 3) Urychlenou deepoxidací pigmentů xantofylového cyklu u rostlin A. thaliana a semenáčků smrku, které byly vystaveny kombinovanému stresu z vysoké ozářenosti a zvýšené teploty a 4) Ochrannou úlohou UVA záření proti poškození vyvolanému UVB zářením.

Abstract:

My dissertation deals with the effect of abiotic factors such as a spectral quality of radiation (and its intensity) and elevated temperature on qualitative and quantitative composition of photosynthetic pigments (chlorophylls and carotenoids). These pigments have very important role in the photosynthetic apparatus. They are key pigments not only in absorption of incident radiation, but they are very important in protection of photosynthetic apparatus against photooxidative stress. Most important methods used in experimental work were absorption spectroscopy and high performance liquid chromatography. For identification of chlorophyll derivatives, the Raman and mass spectrometry and electrophoresis (SDS-PAGE, CN-PAGE) were also used. Theoretical part of my dissertation summarizes the importance of photosynthetic pigments of higher plants and their biosynthetic pathways. In the part of thesis dealing with chlorophylls, detailed description of structure and function of enzyme Geranylgeranyl reductase, its relation with functional state of photosynthetic apparatus and cumulation of chlorophyll´s derivatives are included. The second section of theoretical part summarizes information related to the protective role of carotenoids against photooxidative damage, specifically the thermal dissipation of excessive excitation energy and protective function of xanthophyll cycle. Both groups of pigments (its quantity and quality) are studied in connection with above mentioned abiotic stresses, such as spectral quality of light, UV radiation, high light and high temperature. Experimental part of my thesis contains motivation for research and comments to my publications, which are aimed at: 1) Identification of chlorophyll derivatives accumulated in plants cultivated under the green light, 2) Reacclimation of plants transferred from high light conditions to low light and the role of carotenoids and UV-shielding compounds in photosynthetic apparatus protection, 3) Facilitated deepoxidation of xanthophyll cycle pigments of A. thaliana plants and spruce seedlings exposed to the combined effect of high light and elevated temperature, 4) Protective role of UVA radiation against a damage induced by UVB radiation.
