Preparation and functional characterization of recombinant proteins AHP4 and AHP6

Abstract:

Přenos rostlinného cytokininového signálu je zprostředkován tzv. vícekomponentním systémem, který se vyvinul z bakteriálního dvoukomponentního systému. V těchto systémech fungují proteiny AHP jako můstky mezi příjmem signálu receptory v cytoplazmatické membráně a aktivací transkripčních faktorů v jádře. Protein AHP6 je, na rozdíl od ostatních AHP, inhibitorem této dráhy. V poslední době se předpokládá, že i protein AHP4 by mohl být jejím negativním regulátorem. První část této diplomové práce se zabývala optimalizací podmínek pro expresi rekombinantních proteinů AHP4 a AHP6 a optimalizací přípravy těchto proteinů o dostatečné čistotě, aby mohly být použity pro následnou strukturní analýzu. K optimalizaci přípravy proteinů bylo využito purifikačních metod. V druhé části byla s využitím western blotingu testována specifita monoklonálních protilátek připravených proti AHP4 a AHP6 k jednotlivým proteinům AHP. Jejich specifita byla testována v lyzátu ze suspenzní kultury A. thaliana, v lyzátu bakterií E. coli a v lyzátu kvasinek Sac. cerevisiae. Byly vybrány protilátky vhodné k dalšímu použití.

Abstract:

Cytokinin signal transduction in plant cells is mediated via the so-called multistep phosphorelay, which had developed from the bacterial two-component system. In these arrangements, AHP proteins are known to function as connectors between the income signal (via the cytoplasmic membrane receptors) and the transcription factors activation in nucleus. The AHP6 protein is an inhibitory agent of this pathway, contrary to the other AHP proteins. Recently it has been thought that either the AHP4 protein might manage to be inhibitory. The first part of the diploma thesis is dealing with the optimization of expression conditions for recombinant AHP4 and AHP6 proteins. Preparation step in order to get the proteins of a maximum purity possible for the structure analysis was being further optimized. In the second part, specifity of the monoclonal anti AHP4 and anti AHP6 was tested with the use of western blot. The specifity of the antibodies was examined using the lysates of the suspense cultures of A. thaliana, E. coli a Sac. cerevisiae. The proper antibodies for following experiments were chosen.
