The role of AGR2 and AGR3 proteins in cancer cell in response to stress conditions

Abstract:

Jedním z nově studovaných mechanismů regulace genové exprese nádorových onkogenů jsou mechanismy spojené s nekódujícími oblastmi mRNA, tzv. nepřekládanými oblastmi (UTR, untranslated regions). Zatímco 5´UTR ovlivňuje především translaci, 3´UTR oblast je zodpovědná za stabilitu mRNA, její buněčnou lokalizaci a stejně jako 5´UTR reguluje efektivitu translace. Obě oblasti obsahují řadu regulačních sekvencí, ať už s inhibičním nebo stimulačním efektem. Během evoluce či v důsledku dalších biologických procesů často dochází ke ztrátám těchto nekódujících oblastí a s tím jsou následně spojeny změny expresních profilů dotčených genů. Studium regulace exprese se v současnosti zaměřuje především na nekódující oblast 3´UTR, na které je lokalizována řada inhibičních regulačních míst. Změny v délce 3´UTR, v nukleotidové sekvenci, možná translokace 3´UTR v rámci transkriptomu nebo úplná ztráta 3´UTR umožňují tvorbu specifických sestřihových variant mRNA lišících se navzájem nejen délkou a přítomností 3´UTR, ale také možným C-koncovým zkrácením kódující sekvence. Tato variabilita mRNA může výrazně ovlivňovat genovou expresi onkogenů vedoucí až ke změně buněčných charakteristik jako je proliferace, přežití nebo migrace a invazivita. U řady onkogenů byly tyto změny v jejich mRNA již popsány, obvykle jsou spojeny s jejich zvýšenou expresí, čímž přispívají k rozvoji maligního fenotypu. Tato práce je zaměřená na objasnění mechanismu regulace genové exprese nekódující regulační oblastí 3´UTR nově popsaného nádorového markeru genu Anterior Gradient 2 (AGR2), který kóduje protein s prokazatelnými pro-onkogenními vlastnostmi a jehož zvýšená exprese je spojována s různými typy nádorových onemocnění. Vlastní regulace exprese AGR2 doposud není objasněna a postupně je zkoumána úloha AGR2 ve vnitrobuněčných signálních drahách spojených s maligní transformací. V první části práce byly in silico analyzovány sekvenční rozdíly sestřihových variant AGR2-001 a AGR2-005 lišících se právě C-koncovou oblastí kódující sekvence a UTR oblastmi. Následně byl stanoven rozdílný expresní profil obou variant AGR2, který je závislý na přítomnosti příslušných UTR oblastí. Studium mRNA AGR2 ukázalo inhibiční vliv délky 3´UTR na expresi AGR2 – čím je 3´UTR delší, tím nižší je exprese. Následující in silico analýzou byly predikovány různé regulační sekvence, především v distální části 3´UTR mRNA AGR2, které mohou ovlivňovat délku 3´UTR a v důsledku toho i hladinu proteinu. V další části práce byla s využitím qRT-PCR identifikována zkrácená délka 3´UTR mRNA AGR2 na panelu nádorových buněčných linií s endogenní expresí AGR2. Zkrácená délka 3´UTR byla pozorována také v klinických vzorcích nádorových tkání karcinomu prsu se zvýšenou expresí AGR2, přičemž výraznější zkrácení korelovalo se silnější expresí AGR2. Na panelu vzorků nádorových tkání s detekovanou expresí AGR2 bylo pozorováno výraznější zkrácení 3´UTR mRNA AGR2 ve srovnání se vzorky příslušné nenádorové tkáně. V poslední části práce byl objasněn jeden z molekulárních mechanismů regulace exprese AGR2. Aktivací mTORC1 dochází ke zkracování 3´UTR mRNA AGR2, což má za následek zvýšení hladiny exprese AGR2.

Abstract:

One of the newly studied mechanisms of regulation of gene expression of tumor oncogenes are mechanisms associated with untranslated regions (UTRs) of mRNA. While 5´UTR affects primarily translation, the 3´UTR region is responsible for the mRNA stability, mRNA cell localization and, as well as 5´UTR, it is involved in regulation of translation efficiency. Both regions contain a number of inhibiting or stimulating regulatory sequences. During evolution or as a result of other biological processes, these non-coding regions are often lost, and consequently changes in the expression profiles of the genes are associated Expression regulation is currently focused on the non-coding region 3´UTR, on which a number of inhibitory regulatory sites are localized. Changes in the 3´UTR length, in the nucleotide sequence, possible 3´UTR translocations within the transcriptome or the complete 3´UTR loss, allow for the generation of multiple specific splice variants of mRNA differing not only in the length and presence of 3´UTR, but also in the possible C-terminal truncation of coding sequence. This variability of mRNA can significantly affect gene expression of oncogenes leading to changes in cellular characteristics such as proliferation, survival or migration and invasivity. These changes in their mRNA have already been recognized for more oncogenes. They are usually associated with their increased expression and thus contribute to the development of a malignant phenotype. This work is focused on evaluation of the regulation mechanism of gene expression influenced by non-coding regulatory region 3´UTR of the newly described tumour marker gene Anterior Gradient 2 (AGR2) encoding protein with demonstrable pro-oncogenic properties and increased expression is associated with various tumour diseases. The actual regulation of AGR2 expression has not yet been elucidated and the role of AGR2 in intracellular signalling pathways is being studied. In the first part of the thesis, the sequence variance of the splice variants AGR2-001 and AGR2-005 differing in the C-terminal region of the coding sequence and the UTR regions were analysed in silico. Subsequently, a different expression profile of both AGR2 variants, dependent on the presence of the respective UTR regions, was determined. Study of AGR2 mRNA showed the inhibitory effect of 3´UTR on the expression of AGR2 – the longer 3´UTR is, the weaker the expression is. Following in silico analysis, various regulatory sequences, particularly in the distal part on the 3´UTR mRNA of AGR2 were predicted. They may influence the length of 3´UTR and, consequently, the protein level. In the following part of the thesis, a shortened length of 3´UTR mRNA of AGR2 on a panel of tumour cell lines with endogenous expression of AGR2 was identified using qRT-PCR. The shortened length of 3´UTR was also observed within the breast tumour tissue samples with increased AGR2 expression, where more significant truncations correlated with stronger AGR2 expression. Using samples of tumour breast tissues with detected AGR2 expression, more pronounced truncation of 3´UTR mRNA of AGR2 was observed in comparison to samples of the respective non-tumour tissues. In the last part of the thesis, one of the molecular mechanisms of regulation of AGR2 expression was elucidated. Activation of mTORC1 reduces the 3´UTR mRNA of AGR2, resulting in increased levels of AGR2 expression.
