Abstract:
Tato diplomová práce se zabývá využitím funkčního testu proteinu Atm pro detekci mutací u pacientů s chronickou lymfocytární leukémií. Test je založen na sledování indukce genu CDKN1A (p21) po paralelním ošetření buněk fludarabinem a doxorubicinem, kdy nefunkční protein Atm narušuje indukci p21 po doxorubicinu. Abstract:
The thesis dealt with the functional test of Atm protein for ATM mutations´ detection in patients with chronic lymphocytic leukemia. The test is based on the evaluation of CDKN1A (p21) gene induction after parallel treatment of cells with fludarabine and doxorubicin; dysfunctional Atm protein impairs p21 induction after doxorubicin administration.
