Characterisation of telomere- and telomerase-associated proteins

Abstract:

Telomeres are specialized structures at the end of eukaryotic chromosomes. The enzyme telomerase – reverse transcriptase, is involved in maintaining their length, which is important for protecting the ends of linear chromosomes. It consists of two major subunits: telomerase RNA carrying a template (TR) and telomerase reverse transcriptase (TERT). However, telomerase itself does not consist of only these two subunits - other proteins interacting with both subunits or proteins regulating telomerase access to telomeres are also important components. Protein complexes performing this function have been described in detail in yeasts or animals. In mammals, for example, the Shelterin protein complex is known. A similar complex has not yet been identified in plants, but candidate proteins with similar functions as Shelterin components are known. This diploma thesis deals with the study of plant-specific proteins from the SMH (Single-Myb-Histone) group, TRB proteins. In the Arabidopsis thaliana plant, these proteins are known to be localized on telomeres, to be able to form dimers, and even to interact with the AtTERT subunit of telomerase. Three PpTRB proteins have been identified in the Physcomitrella patens moss, but have not yet been further characterized. It is therefore questionable whether the function of these proteins is evolutionarily conserved. The aim of this work was to determine whether PpTRB proteins have similar interaction partners as AtTRB proteins, and thus, whether they interact with each other or with the PpTERT subunit of telomerase.

Abstract:

Teloméry sú špecializované štruktúry nachádzajúce sa na koncoch eukaryotických chromozómov. Na udržiavaní ich dĺžky, ktorá je dôležitá pre ochranu koncov lineárnych chromozómov, sa podieľa enzým telomeráza – reverzná transkriptáza. Tento enzým je tvorený dvomi hlavnými podjednotkami: telomerázovou RNA nesúcou templát (TR) a telomerázovou reverznou transkriptázou (TERT). Samotnú telomerázu však netvoria len tieto dve podjednotky – dôležitou súčasťou sú aj ďalšie proteíny interagujúce s oboma podjednotkami alebo proteíny, ktoré môžu regulovať prístup telomerázy k teloméram. Proteínové komplexy vykonávajúce túto funkciu boli podrobne opísané u kvasiniek či živočíchov. U cicavcov je napríklad známy proteínový komplex Shelterin. U rastlín zatiaľ nebol identifikovaný podobný komplex, sú však známe kandidátne proteíny spĺňajúce podobné funkcie ako zložky Shelterinu. Táto diplomová práca sa zaoberá štúdiom rastlinne-špecifických proteínov zo SMH (Single-Myb-Histone) skupiny, TRB proteínov. U rastliny Arabidopsis thaliana je o týchto proteínoch známe, že sú lokalizované na telomérach, sú schopné tvoriť diméry a dokonca interagujú s AtTERT podjednotkou telomerázy. U machu Physcomitrella patens boli identifikované tri PpTRB proteíny, dosiaľ však neboli bližšie charakterizované. Je teda otázne, či je funkcia týchto proteínov evolučne konzervovaná . Cieľom tejto práce bolo zistiť, či majú PpTRB proteíny podobných interakčných partnerov ako AtTRB proteíny, a teda či interagujú vzájomne medzi sebou či s PpTERT podjednotkou telomerázy.
