Modulation of specific immune response and hypersensitivity reaction by pseurotins

Abstract:

Deregulovaná kontrola imunitního systému má velké zdravotní důsledky a stojí za rozvojem řady poruch, mezi které patří autoimunitní a alergická onemocnění. V současné době frekvence alergických onemocnění neustále stoupá, což přináší potřebu zlepšit naše chápání molekulárního mechanismu za těmito patologiemi a také aktivní hledání nového terapeutického přístupu. Důležitou inspirací pro vývoj léčiv jsou bioaktivní přírodní sloučeniny, které mohou ovlivnit specifické buněčné procesy a zároveň mají potenciál stát se terapeuticky využitelnými. Mezi těmito sloučeninami jsou i sekundární metabolity hub ze skupiny pseurotinů. Specifická modulační aktivita pseurotinů na imunitní buňky však není dobře známa. Hlavním cílem mé práce bylo popsat mechanismy hypersenzitivní reakce opožděného typu indukované kuřecím ovalbuminem in vivo a také kompletní analýza účinků přírodních pseurotinů a jejich syntetických analogů na T a B buňky, jako hlavní buňky specifického imunitní odpovědi. Nejprve byly provedeny testy in vitro, konkrétně byl testován účinek přírodního pseurotinu D na izolované T buňky. Naivní T lymfocyty byly izolovány z myších lymfatických uzlin, aktivovány a diferencovány na Th1, Th2 subpopulace. Pseurotin D zprostředkoval snížení proliferace a diferenciace Th1 a Th2 buněk, jak bylo vyvozeno na základě inhibice genové exprese Gata3 a Tbx21 a produkce efektorových cytokinů interferonu γ a interleukinu 13. Kromě toho byl in vitro testován účinek pseurotinu D na proliferaci aktivovaných B buněk a diferenciaci na plazmatické buňky. Významná inhibice produkce imunoglobulinu E a M korelovala se zastavením buněčné proliferace a diferenciace. Komplexní analýza ovlivnění intracelulárních signálních drah odhalila, že účinky pseurotinů souvisí s inhibicí signální dráhy JAK/STAT. Analýza syntetických analogů pseurotinu ukázala podobný nebo dokonce téměř o jeden řád vyšší potenciál inhibovat aktivaci B buněk. Nakonec byly hodnoceny účinky pseurotinu D na zpoždění reakce přecitlivělosti typu IV na myším modelu. Byl analyzován účinek pseurotinu intravenózně aplikovaného před druhou posilovací dávkou ovalbuminu podávaného senzibilizovaným myším. Pseurotin D významně snížil otok tlapek, hlavní příznak reakce přecitlivělosti typu IV v tkáni, stejně jako produkci prozánětlivých a protizánětlivých cytokinů v tkáni tlapek a zvětšení sleziny. Celkově výsledky shrnuté v této práci jasně ukazují, že přírodní pseurotiny a jejich syntetické analogy mohou snižovat specifickou imunitní odpověď. Pseurotiny by tedy mohly být navrženy jako potenciální léčiva pro léčbu onemocnění u kterých je základem patologicky deregulovaná imunitní odpověď, například alergií

Abstract:

Dysregulated control of the immune system has major health consequences and is behind the development of numerous disorders that include autoimmune and allergic diseases. Currently, the frequency of allergic diseases is constantly rising, bringing a need to improve our understanding of the molecular mechanism behind these pathologies, as well as the active search for a new therapeutic approach. Important inspiration for drug development is bioactive natural compounds that can affect specific cellular processes and, at the same time, have a potential to become therapeutically applicable. Among these compounds are secondary fungi metabolites from the pseurotin family. However, the specific modulatory activity of pseurotins on immune cells is not well understood. The main aim of my work was to describe the mechanisms of delayed-type hypersensitivity reaction induced by chicken ovalbumin in vivo, as well as to complete analysis of effects o natural pseurotins and their synthetic analogues on main cells of the specific immune response, T and B cells. Initially, in vitro tests were performed, namely, the effect of natural pseurotin D on isolated T cells was tested. Naive T lymphocytes were isolated from mouse lymph nodes, activated, and differentiated into Th1, Th2 subpopulations. Pseurotin D mediated a decrease in proliferation and differentiation of Th1 and Th2 cells, as was concluded based on inhibition of the gene expressions of Gata3 and Tbx21 and the production of the effector cytokines interferon γ and interleukin 13. In addition, the effect of pseurotin D on activated B cell proliferation and differentiation into plasma cells was tested in vitro. Significant inhibition of IgE and IgM production was correlated with the arrest of cell proliferation and differentiation. Comprehensive analysis of affected intracellular signaling pathways revealed that effects of pseurotins are related to inhibition of the JAK / STAT signaling pathway. Analysis of synthetic pseurotin analogs showed similar or even almost one order higher potential to inhibit B cell activation. Finally, the effects of pseurotin D on the delay in type IV hypersensitivity response were evaluated. Intravenously applied pseurotin before second booster of chicken ovalbumin to mice sensitized with ovalbumin was analysed. Pseurotin D significantly decreased paw swelling, the main symptom of type IV hypersensitivity response in a tissue as well as production of pro-inflammatory and anti-inflammatory cytokines in the tissue of the paws and spleen enlargement. Overall, the results summarized in this thesis clearly show that natural pseurotins and their synthetic analogues can down-regulate specific immune response. Thus, pseurotins could be recognised as potential drugs for the treatment of diseases with a base in pathologically deregulated immune response such as allergies.
