Determination of riboflavin in beer using CELIF

Abstract:

V teoretické části bakalářské práce byl popsán význam riboflavinu a jeho zdroje. Teoretická část se věnovala také principu metody CE-LIF a jejím praktickým využitím. Prvním cílem praktické části bylo stanovení citlivosti metody; ta byla ověřena na roztoku standardu riboflavinu. Reprodukovatelnost měření standardu ve fosfátovém pufru o koncentraci 30 mM a pH 9,1 byla uspokojivá. Druhým cílem bylo ověření přítomnosti riboflavinu v pivu a zvolení optimálních podmínek pro další analýzu vzorků. Třetím prioritním cílem experimentu bylo zjistit u různých druhů piv jejich koncentrace riboflavinu. Nejmenší koncentrace byla zjištěna u piva Medium, Starobrno a nejvyšší obsah riboflavinu má ležák Staropramen. Základní cíl, tj. ověření, zda je možné využít běžnou metodu CE – LIF s levným diodovým laserem 405 nm. Stanovení riboflavinu v pivu byl splněn, použití CE – LIF je možné. Vzorek piva (Ležák, Starobrno) byl pro srovnání i změřen přístrojem fluorimetricky kvůli posouzení citlivosti metod. i přístrojem fluorimetrem pro srovnání, která metoda je citlivější. Koncentrace riboflavinu v pivu u metody fluorimetru byla zjištěna 6,4•10-3 mg/l a u metody CE – LIF 3,4 mg/l. CE-LIF metoda je citlivější než metoda fluorimetrie. Metoda standardního přídavku u fluorimetrického stanovení nevedla k dosažení očekávaného výsledku, vliv matric pravděpodobně nebyl odstraněn. Pro zajímavost byl proveden výpočet, kolik by se muselo vypít piv, aby se dosáhla doporučená denní dávka vitaminu B2, která se pohybuje kolem 1,1 – 1,3 mg•den-1. Muselo by se vypít 441,8 ml piva.

Abstract:

The theoretical part of the thesis clarified the significance of riboflavin and its resources. Furthermore, it dealt with the principle of CE-LIF method and its practical use. The first objective was to determine the practical sensitivity of the method, which was tested on the standard solution of riboflavin. The reproducibility of the measurement of the standard in phosphate buffer at a concentration of 30 mM and pH 9.1 was satisfactory. The second objective was to verify the presence of riboflavin in beer and to select optimum conditions for further analysis of samples. The third priority objective of the experiment was to determine the concentration of riboflavin in different types of beer. The lowest concentration was found in the beer Medium made by Starobrno and the highest riboflavin content was detected in the lager made by Staropramen. The main goal, i.e. verification of the possibility to use the ordinary CE-LIF method with low-cost diode lasers at 405 nm to determine the presence of riboflavin, was fulfilled: it is possible to use the CE-LIF method. The conclusion is that the device CE - LIF is suitable for the separation of riboflavin in beer. Determination of riboflavin in beer was achieved, the use of CE – LIF is possible. The beer samples were also measured by a fluorometer device in order to compare the sensitivity of the two methods. The concentration of riboflavin in beer was found 6.4•10-3 mg/l when measured by the fluorometer and 3.4 mg/l when measured by the CE-LIF method. The method of standard addition in fluorometric determination did not yield the anticipated result; the influence of matrices was probably not removed. Just as a matter of interest, it was also calculated how much beer one would have to drink to reach the recommended daily dose of vitamin B2, which is around 1.1 to 1.3 mg a day-1. One would have to drink 441,8 mililiters of beer.
