Use of a selective stimuli-responsive fluorescent probe for the determination of metallothionein by capillary electrophoresis

Abstract:

V dnešní době je vědecké zaměření na metalothioneiny (MTs) stále větší a více se dostává do popředí. U těchto proteinů totiž byla prokázaná schopnost chránit nádorové buňky proti protinádorovým léčivům. MTs se účastní řady důležitých obranných reakcí v organismu, zejména při stresových situacích. Hlavní biologická role těchto proteinů zatím zůstává tajemstvím, je ale zřejmé, že metalothioneiny mají mnohostranné funkce a jejich důležitost je závislá na specifických požadavcích každého konkrétního organismu. Tato práce se zaměřuje na detekci MTs pomocí selektivní fluorescenční sondy a kapilární elektroforézy s laserem indukovanou fluorescencí. Výsledky potvrzují selektivitu sondy a její vhodnost pro citlivé stanovení MTs v biologických vzorcích. Mezi vize do budoucna patří výzkum přesnějšího fungování těchto proteinů, nebo pro využití monitorování stavu životního prostředí.

Abstract:

Nowadays, the scientific focus on metallothioneins (MTs) is getting bigger and more prominent. These proteins have been shown to protect tumor cells against anticarcinogenic drugs. MTs participate in many important defense reactions in the organism, especially in stressful situations. The main biological role of these proteins still remains uncovered, but it is clear that metallothioneins have multifaceted functions and their importance depends on the specific requirements of each specific organism. This work focuses on the analysis of MTs using a selective fluorescent probe and capillary electrophoresis with laser-induced fluorescence. The results confirm the selectivity of the probe and its suitability for sensitive determination of MTs in biological samples. Visions for the future include research into the more precise functioning of these proteins, or for use in environmental monitoring.
