Deletion of SHOX gene in patients with growth failure

Abstract:

Bakalářská práce je studií provedenou u 23 pacientů s podezřením na mikrodeleci oblasti chromozomu X zahrnující SHOX gen. Základním kritériem pro výběr pacientů byla přítomnost poruchy růstu. S větší četností se ve vybraném souboru pacientů vyskytly i symptomy jako opožděný kostní věk, faciální stigmatizace a níže posazené ušní boltce. Léri-Weill dyschonrosteóza nebo Madelungova deformita, typické projevy této mikrodelece, byla přítomna pouze u třech pacientů. K detekci haploinsuficience genu SHOX byla použita metoda fluorescenční in situ hybridizace s použitím sond POSEIDONTM firmy KREATECH. Touto metodou bylo v analyzovaném souboru pacientů odhaleno 5 delecí SHOX genu (22%). Ve třech případech šlo o rodinný výskyt mikrodelece SHOX genu s normálním mužským či ženským karyotypem. U dvou pacientů byla odhalena haploinsuficience SHOX genu jako důsledek rozsáhlejší chromozomové aberace mos 45,X/46,X, psuidic(Y)(q11.22) a 46,X,del(X)(p22.33).

Abstract:

My bachelor´s work has been a study made on 23 patients suspected of microdeletion of X chromosome region extending SHOX gene. The basic criterion for patients selection had been an occurence of growth disorder. Symptoms like delayed bone age, facial stigmatization and lower perched auricles were more frequent in the selected aggregate of patients. Léri-Weill dyschondrosteosis or Madelung´s deformity, typical displays of the microdeletion, had only three patients. Fluorescence in situ hybridization method with POSEIDONTM probe by KREATECH company had been used to detect haploinsuficiency of SHOX gene. This method detected 5 deletion of SHOX gene (22%) in the analyzed aggregate of patients. In three cases there was a family occurence of SHOX gene´s microdeletion with normal male or female karyotype. The haploinsuficiency of SHOX gene as a consequence of more extensive chromosome aberation was revealed at 2 patients - mos 45,X/46,X, psuidic(Y)(q11.22) and 46,X,del(X)(p22.33).
