Epigenetics features of mouse embryonic stem cells diferentiated into cadiomyocytec

Abstract:

Embryonální kmenové buňky (ESC) jsou pluripotentní buňky schopné víceliniové diferenciace. Nyní je známo, že epigenetické procesy jsou zásadní pro pluripotentní rysy ESC. Přesná genová exprese, programy buněčného osudu nebo reakce na buněčný stres vyžadují harmonickou souhru mezi všemi epigenetickými regulačními úrovněmi. Nedávné zkoumání post-transkripčních modifikací RNA však rozšířilo pole epigenetiky a zvláštní methylace v poloze N6 adenosinu, nazývaná m6A RNA. Tento objev získal velkou pozornost kvůli své kritické roli při regulaci zpracování RNA.V této práci byla zkoumána role různých epigenetických markerů nebo modifikátorů pomocí explantovaných embryonálních srdcí nebo diferenciačního modelu myších embryonálních kmenových buněk (mESC) se zvláštním zaměřením na diferencované kardiomyocyty. Pro tento účel byly použity dvě myší ESC linie: mESC divokého typu (wt) a mESC, které jsou deficitní pro histon deacetylázu 1 (HDAC1). Navíc bylo dosaženo experimentálně indukovaných změn v posttranslačních modifikacích histonů použitím tří různých inhibitorů histonových deacetyláz (HDACis). První část práce analyzuje tepající fenotyp diferencovaných kardiomyocytů v závislosti na deficitu HDAC1 nebo diferencující kardiomyocyty ovlivněné HDAC inhibitory. Na základě tohoto experimentálního přístupu byly studovány vybrané histonové značky u diferencovaných mESC (wt nebo HDAC1 deficitních, dn) nebo u explantovaných embryonálních srdcí. Ve druhé části byla pomocí zavedených diferenciačních modelů (mESC wt a HDAC1 dn) nebo explantovaných embryonálních srdcí studována lokalizace a profily modifikátorů m6A RNA (METLL 3, METTL 14, METTL 16 a FTO) v průběhu diferenciace a u terminálně diferencovaných kardiomyocytů. Kromě toho, s ohledem na rostoucí důkazy o křížové komunikaci mezi regulací epigenomu a epitrascriptomu, byl zkoumán i možný vliv deplece HDAC1 nebo ošetření HDACis na m6A RNA a její modifikátory v různých typech RNA

Abstract:

Embryonic stem cells (ESC) are pluripotent cells capable of multilineage differentiation. Epigenetic processes are now known to be essential for the pluripotent features of ESC. The accurate gene expression, cell fate programs, or cell stress response requires the harmonious interplay between all epigenetic regulatory layers. However, recent exploration of post-transcriptional modifications of RNA has broadened the field of epigenetics, and particular methylation at the N6 position of adenosine, termed m6A RNA, has attracted the most attention because of its critical role in regulating RNA processing. In the present thesis, the role of different epigenetic markers or modifiers was explored using explanted embryonic hearts or a mouse embryonic stem cells (mESC) differentiation model, with a particular focus on differentiated-cardiomyocytes. For this purpose, two mESC lines were used: mESCs wild-type (wt) and mESCs deficient of histone deacetylase 1 (HDAC1 dn). Moreover, experimentally induced changes in histone post-translational modifications were achieved by employing three different histone deacetylase inhibitors (HDACis). The first part analyzes the beating phenotype of differentiated cardiomyocytes depending on HDAC1 deficiency or differentiating cardiomyocytes affected by HDAC inhibitors. Based on this experimental approach, selected histone markers were studied in differentiated mESCs (wt or HDAC1 deficient, dn) or in explanted embryonic hearts. In the second part, using the established differentiation models (wt or HDAC1 deficient) or explanted embryonic heart, the localization and profile of the m6A RNA modifiers (METLL 3, METTL 14, METTL 16, and FTO) were studied over the differentiation and in differentiated-cardiomyocytes. Moreover, considering the growing evidence of a cross-talk between epigenome and epitrascriptome regulation, the possible influence of HDAC1 depletion or HDACis treatment on m6A RNA and its modifiers was investigated in different types of RNA.
