Effect of flavonoids on expression of genes involved in oxidative stress response

Abstract:

Oxidační stres je diskutovaným tématem dnešní doby, neboť jsme stále více obklopeni chemickými látkami a škodlivými vlivy, které mohou negativně působit na naše zdraví. Tato práce se zaměřuje na geny zapojené v obraně proti oxidačnímu stresu ovlivněné flavonoidy a dále vyhodnocuje vliv flavonoidních látek na expresi genů za přítomnosti toxických látek nebo ionizujícího záření. Práce představuje shrnutí několika studií, které tuto problematiku sledovaly a pro její objasnění použily různé metody. Pro stanovení exprese genů se nejčastěji využívaná metoda qPCR. Hlavním přínosem této práce je zjištění, že flavonoidní látky zvyšují aktivitu antioxidačních enzymů i zabraňují vzniku oxidačního stresu neutralizací volných radikálů. Míra účinnosti se však u jednotlivých typů flavonoidů liší, a to jak v reakci na konkrétní toxickou látku, použité ionizující záření nebo vyčerpání GHS.

Abstract:

Oxidative stress is a hotly debated topic today as we are increasingly surrounded by chemicals and harmful influences that can negatively affect our health. This work focuses on genes involved in the defense against oxidative stress affected by flavonoids and further evaluates the effect of flavonoid compounds on gene expression in the presence of toxic substances or ionizing radiation. This paper presents a summary of several studies that have investigated this issue and used different methods to elucidate it. The most commonly used method for gene expression determination is qPCR. The main contribution of this work is the finding that flavonoid substances increase the activity of antioxidant enzymes as well as prevent oxidative stress by neutralizing free radicals. However, the degree of efficacy varies among flavonoid types in response to the specific toxicant, ionizing radiation used, or GHS depletion.
