Testing the susceptibility/resistance of transgenic barley lines expressing antimicrobial peptide during the pathogenesis of phytopathogenic fungi from the genus Fusarium

Abstract:

Antimikrobiální peptidy jsou důležitou složkou vrozeného imunitního systému téměř všech živých organismů. Tyto nízkomolekulární peptidy jsou považovány za prospěšné terapeutické látky díky jejich vysoké selektivitě a jedinečnému způsobu působení, který brání vzniku rezistentních kmenů patogenů. Jsou tak vhodnou alternativou pro vývoj léčiv nové generace. Kromě terapeutického průmyslu mohou být využity k ochraně rostlin v zemědělství a pomoci tak překonat ztráty ve výnosu a kvalitě zemědělských produktů. Mezi nejzávažnější choroby obilovin patří fuzariózy klasů, které z důvodu produkce mykotoxinů způsobují velké hospodářské ztráty. Tato bakalářská práce je zaměřena na analýzu rostlin ječmene (Hordeum vulgare L., cv. Golden Promise) exprimujících derivát lidského -defensinu 2. Na genomové úrovni byly vyselektovány transgenní linie T3 generace. Na základě relativní kvantifikace exprese transgenu DEF byla ověřena funkčnost a zrnová specifita globulinového promotoru u klíčících zrn ječmene. U ječmene byl dále sledován fenotyp a množství biomasy při napadení fytopatogenní houbou rodu Fusarium graminearum.

Abstract:

Antimicrobial peptides are important components of the innate immune system of nearly all living organisms. These low molecular mass peptides are considered as prospective therapeutic agents due to their high selectivity and unique mode of action that prevents the emergence of resistant pathogenic strains. In addition to the therapeutic industry antimicrobial peptides can also be employed in agriculture for plant protection and thus help surmount losses in yield and quality of agricultural products. Fusarium head blight disease belongs among the most serious disease of cereals, which due to the production causing major economic losses. This bachelor thesis is focused on analysis of barley plants (Hordeum vulgare L., cv. Golden Promise) expressing derivative of human -defensin 2. Independently transformed transgenic barley T3 lines were selected at the genome level. Functionality and grain specifity of the globulin promoter were confirmed with relative quantificiation of DEF transgene expression in germinating grains. Phenotype and dry weight quantification of biomass were monitored in the barley during infection with the phytopatogenic fungus Fusarium graminearum.
