Motility of eugregarines (Apicomplexa) in vitro and in vivo

Abstract:

Gregariny, patřící mezi bazální výtrusovce (Apicomplexa), představují velice různorodou skupinu eukaryotických jednobuněčných organismů. Parazitují v tělní dutině a ve vnitřních orgánech bezobratlých živočichů obývajících terestrické i vodní prostředí, přičemž typickými hostiteli jsou zejména členovci a kroužkovci. Eugregariny se vyskytují hojně i u laboratorně chovaného hmyzu, a tak představují snadno dostupný a finančně nenáročný model pro různé experimentální přístupy. Přesto jim však nebyla věnována dostatečná pozornost a jejich biologie není důkladně prozkoumána. Vzhledem k tomu, že výtrusovci zahrnují řadu významných patogenů způsobujících závažná onemocnění lidí i hospodářských zvířat (např. malárie, toxoplazmóza, kryptosporidióza), důkladné prozkoumání zástupců jejich bazálních linií by mohlo pomoci odhalit nové strategie v boji proti medicínsky i ekonomicky významným druhům. Cílem práce je navrhnout a otestovat in vitro a in vivo postupy vhodné ke studiu pohybu u modelových druhů eugregarin s následnou cytologickou a histochemickou analýzou buněčných struktur zapojených do jejich pohybových mechanismů. Rešeršní část se zabývá obecnou charakteristikou výtrusovců a sumarizuje základní poznatky o apikálním komplexu a dalších strukturách zapojených do pohybu těchto organismů. Dále se věnuje popisu hlavních pohybových struktur eugregarin a sumarizuje jejich životní cyklus.

Abstract:

Gregarines, belonging to the basal Apicomplexa, represent a highly diverse group of eukaryotic unicellular organisms. They parasitize the body cavity and internal organs of invertebrates inhabiting terrestrial and aquatic environments, with typical hosts being arthropods and annelids. Since eugregarines are also abundant in laboratory insect colonies, they represent an accessible and financially available model for various experimental approaches. Despite this, however, not enough attention is paid to them, and their biology remains poorly investigated. Given that apicomplexans include several pathogens responsible for serious human and livestock diseases (e.g., malaria, toxoplasmosis, cryptosporidiosis), a thorough research on the basal lineage representatives could help develop new strategies to combat medically and economically important species. The aim of the thesis is to design and test in vitro and in vivo procedures suitable for the study of motility in model eugregarine species with subsequent cytological and histochemical analysis of cell structures involved in their movement mechanisms. The theoretical part of the thesis deals with the general characteristics of Apicomplexa and summarizes the knowledge about the apical complex and other structures involved in the apicomplexan motility. This part further describes the major motility structures of eugregarines and summarises their life cycle.
