Fotoochranné mechanismy mutantů Arabidpsis thaliana s modifikovanou strukturou PS II a limitací xantofylového cyklu

Photoprotective mechanisms in Arabidopsis thaliana mutants with modified PS II structure and limitations of the xanthophyll cycle

Abstract:

V současné době jsou procesy fotosyntézy intenzivně studovány, zejména v oblasti stresové fyziologie rostlin. Byla vyvinuta řada biochemických a biofyzikálních metod, které umožňují sledovat tyto procesy s velkou přesností. Experimenty v této práci byly zaměřeny na fotochemické procesy ve fotosystému II a na xantofylový cyklus. Pro experimenty byly použity rostliny Arabidopsis thaliana. V této práci jsem pracoval s lhc mutanty A. thaliana, kteří mají snížené množství světlosběrných komplexů ve fotosystému II a s npq mutanty, kteří mají zablokovaný xantofylový cyklus. Cílem první skupiny experimentů bylo zjistit, jak dokáže fotosyntetický aparát mutantních rostlin fyziologicky reagovat na vysoké dávky fotosynteticky aktivního záření. Cílem druhé skupiny experimentů bylo zjistit vliv účinných látek pesticidů (Previcur a Zolone), které se často používají při kultivaci rostlin, na fotosystém II a xantofylový cyklus. V případě první skupiny experimentů byly rostliny krátkodobě ovlivněny dvěma různými hodnotami fotosynteticky aktivního záření s různou dobou expozice tak, aby celková dávka zářivé energie byla stejná. Tyto experimentální podmínky simulovaly různou radiační zátěž (malou: LL, velkou HL) rostlin A. thaliana. Jako biomarkery fotoinhibičního stresu byly měřeny hodnoty kapacity fotochemických procesů ve fotosystému II (základní fluorescenční poměr Fv/Fm) a byly odebírány a posléze analyzovány vzorky pro stanovení pigmentů pomocí HPLC. Bylo zjištěno množství fotosyntetických pigmentů, pigmentů xantofylového cyklu, jejich deepoxidačního stavu (DEPS) těsně před vystavením experimentálním podmínkám, ihned poté a následně v intervalech 1 hodina, 5 a 12 hodin po ukončení fotoinhibičního ovlivnění. Druhá skupina experimentů studovala vliv chemického ošetření insekticidem (Zolone) a fungicidem (Previcur) v kombinaci s vysokou ozářeností a teplotou na funkci fotosystému II, dále na kapacitu fotoochranných mechanismů. Byly měřeny parametry Fv/Fm, NPQ a odebírány vzorky rostlin pro biochemické stanovení obsahu fotosyntetických pigmentů, pigmentů xantofylového cyklu, a parametrů DEPS. Z výsledků první skupiny experimentů vyplývá, že nejvyšší senzitivitu vůči fotoinhibici vykazuje mutant lhcb 2-12, u kterého nastal největší pokles parametrů Fv/Fm. Dalším senzitivním mutantem je mutant npq1. Znamená to, že snížené množství světlosběrných komplexů vede ke snížení funkčnosti fotosystému II a vyšší senzitivitě vůči fotosyntetickým zářením indukované fotoinhibici. Výsledky získané u npq mutantů potvrzují klíčovou roli konverze pigmentů xantofylového cyklu v iniciálních fázích fotoinhibice. Ve druhé skupině experimentů u rostlin A. thaliana, které byly ošetřeny insekticidem a fungicidem v kombinaci s experimentálním ovlivněním fotosynteticky aktivního záření a teplotou se prokázalo, že negativní vliv na funkci fotosystému II má Zolone a Zolone v kombinaci s Previcurem. Tento výsledek má význam pro běžnou laboratorní praxi a kultivaci experimentálních rostlin A. thaliana, neboť negativní vliv přípravku Zolone na hodnoty kapacity fotochemických procesů ve fotosystému II je třeba brát v úvahu při kultivacích a následných experimentech zaměřených na různé aspekty fotosyntézy.

Abstract:

Recently, photosynthetic processes are studied intensively, especially in plant stress physiology. Large number of biochemical and biophysical methods have been developed that allow to monitor photosynthetic processes with high accuracy. In this Ph.D. thesis, an attention was devoted to photochemical processes in photosystem II (PSII) and xanthophyll-cycle pigments. For experiements, young plants of Arabidopsis thaliana were used. The measuremets were carried out on lhc mutants that possessed limited amount of light harvesting complexes in PSII antennae and npq mutants that had blocked conversion of violaxanthine to zeaxanthine. The aim of the first group of experiments was to evaluate the responce of A. thaliana mutants to high doses of photosynthetically active radiation (PAR). The aim of the second group of experiments was to find whether or not there is any effect of model fungicide (Previcur) and insecticide (Zolone) on photosynthetic processes in PSII and xanthophyll cycle pigments. In the first experiments, plants were exposed to two different levels of PAR with two different exposition times so that overal light doses were the same. The two levels mimicked different light stress (low light: LL, high light: HL) in A. thaliana plants. Values of the capacity of photochemical processes in PSII (Fv/Fm) vere measured as markers of photoinhibitory stress before and during photoinhbitiory treatments as well as during consequent recovery in dark. Similarly, leaf samples were taken in the same time for furher analysis of de-epoxidation state (DEPS) of xanthophyll-cycle pigments. The second group of experiments studied the effects of Zolone and Previcure in combination with elevated temperature/high light on photosystem II and capacity of photoprotective mechanisms. From the obtained results follows that the highest sensitivity to photoinhibition was found in lhcb2-12 mutant, in which the largest decrease in Fv/Fm values was recorded after photoinhibitory treatment. The second most sensitive A. thaliana plants was npq mutants. It indicated that reduced amount of light harvesting complexes led to a decrease in PSII functioning and increased sensitivity to photoinhibition. The results obtained for npq mutants proved a key role of xanthophyll-cycle pigments conversion in plant photoprotection, especially in the early stages of photoinhibition. It was showed in the second group of experiments with an insecticide/fungicide application combined with high temperature and photoinhibitory treatment, that negative effects on primary photosynthetic processes were caused only by Zolone or Zolone with Previcur. Such results have some general value for common laboratory practice with A. thaliana and its cultivation procedures. The negative effects of Zolone on the capacity of photochemical processes in PSII should be taken into consideration during experimental planning period as well as experiments focused on different aspects of photosynthesis in A. thaliana.
