Enantioselectivity in the uptake of chiral pesticides by earthworms

Abstract:

In this thesis, the fate of pesticides in soil was studied. Tebuconazole, belonging to the group of conazole fungicides,has been chosen as a model pesticide for this work. Tebuconazole is intensively used in agriculture as well as listed as a candidate for substitution due to its persistency and toxicity. Apart from that, there is another interesting aspect of tebuconazole – chirality. Tebuconazole exists in two isomeric forms (R and S enantiomer). Currently, tebuconazole is produced,applied and in majority assessed as a racemic mixture (mixture of both enantiomers in a 1:1 ratio) despite that enantiomers are known to differ in their biological effects. Therefore, our understading of enantiomer-specific behaviour of tebuconazole arising from specific enatiomer-organism interactions requires further attention. The main aim of the theoretical part of this thesis was to define the fate of pesticides in soil, to define chirality and enantioselectivity of conazole fungicides, to define and compare the properties of tebuconazole enantiomers, and at last, to explain the importance of earthworms as model organisms in ecotoxicity testing.The aim of the experimental part of this thesis was to test tebuconazole bioavailability in selected soils (Lufa 2.1, 2.4) by exposing two different species of earthworms (Eisenia adrei, Lumbricus terrestris). This test was carried-out in four variants (Eisenia andrei/Lufa 2.1, Eisenia andrei/Lufa2.4, Lumbricus terrestris/Lufa2.1, Lumbricus terrestris/Lufa2.4) In addition, an elimination test was performed for Lufa 2.4 and Eisenia andrei. The experiments aimed at examining the differences in the uptake and elimination of rac-tebuconazole and its enantiomers (E1, E2) by earthworms. Results revealed that bioaccumulation of rac-tebuconazole was higher in Eisenia andrei compared to Lumbricus terrestris. In all variants, E2 enantiomer showed higher bioaccumulation than E1. The elimination test showed that enantiomer E2 is eliminated from Eisenia andrei more slowly than E1. In light of the above findings, we may conclude that bioaccumulation of tebuconazole was enantiomer specific. This enantioselectivity should be accounted for in the risk assessment of tebuconazole.

Abstract:

V tejto diplomovej práci sa venujeme štúdiu osudu pesticídov v pôdnom prostredí. Modelový pesticíd skúmaný v tejto práci je tebukonazol, patriaci do skupiny konazolových fungicídov, v súčasnosti hojne používaný, ale i tak zaradený medzi kandidátov k substitúcii z dôvodu svojej perzistencie a toxicity. Tebukonazol sa navyše vyznačuje ďalšou významnou vlastnosťou , svojou chiralitou, pričom sa vyrába a do životného prostredia aplikuje v zmesi dvoch izomérov (R a S enantiomér). Tebukonazol v súčasnosti vyrábaný a aplikovaný ako racemická zmes (t.j. zmes dvoch enantiomérov v pomere 1:1) nie je dostatočne skúmaný z hľadiska svojej enantioselektivity u živých organizmov. Pritom je známe, že enantioméry môžu vykazovať rozdielne chovanie a biologické účinky z dôsledku selektívnych interakcii s organizmami v pôdnom prostredí. Cieľom teoretickej časti bolo definovať osud pesticídov v pôdnom prostredí, definovať chiralitu a enantioselektivitu konazolových fungicídov, konkrétne porovnať vlastnosti enantiomérov tebukonazolu a vysvetliť významnosť dážďoviek ako modelových organizmov využívaných v ekotoxikológii.Cieľom experimentálnej časti bolo realizovať dva testy. Najprv sa realizoval test biodostupnosti tebukonazolu voči vybraným druhom dážďoviek Eisenia andrei a Lumbricus terrestris v pôdach Lufa 2.1 a Lufa 2.4 v štyroch variantách (Eisenia andrei/Lufa 2.1, Eisenia andrei/Lufa2.4, Lumbricus terrestris/Lufa 2.1, Lumbricus terrestris/Lufa2.4). Potom sa realizoval test eliminácie za použitia pôdy Lufa 2.4 a dážďovky Eisenia andrei. V týchto testoch sa skúmali rozdiely v príjme a eliminácii rac-tebukonazolu a jeho enantiomérov (E1, E2) dážďovkami. Výsledky testov biodostupnosti preukázali, že bioakumulácia rac-tebukonazolu bola vyššia u druhu Eisenia andrei v porovnaní s Lumbricus terrestris. V prípade každej varianty testu biodostupnosti bola zaznamenaná vyššia bioakumulácia enantioméru E2 do dážďoviek. Výsledky testu eliminácie preukázali , že enantiomér E2 sa z Eisenia andrei eliminoval pomalšie ako enantiomér E1. Z uvedeného plynie, že tebukonazol pri bioakumulácii vykazuje enantioselektívne správanie a táto enantioselektivita by mala byť vzatá do uváženia pri analýze rizík tebukonazolu v prostredí.
