New trends in biomaterial immunocompatibility testing

Abstract:

Význam biomateriálů v biomedicínských aplikacích stále roste, používají se od nosičů léčiv až po tkáňové inženýrství. Pro jejich úspěšné použití je nezbytné pochopení vlastností a interakcí těchto biomateriálů s biologickými systémy. Jedním z kritických aspektů této problematiky je hodnocení reakce imunitního systému, což je v posledních letech předmětem intenzivního výzkumu. K tomu slouží in vitro a in vivo metody, ovšem než se přejde k in vivo testování, musí proběhnout ověření cytotoxicity a imunokompatibility. To se provádí pomocí in vitro metod, které poskytují pohodlné a v rámci možností nákladově efektivní prostředky pro studium reakce imunitního systému na biomateriály. Tato práce se tedy zaměřuje na popis této reakce a metody in vitro používané pro studium imunitní odpovědi na biomateriály na zavedených i méně běžných buněčných populacích. Konkrétně popisuje potenciál makrofágů, neutrofilů, dendritických buněk a lymfocytů pro testování imunokompatibility. Kromě toho tato práce zkoumá využití kokultivací v biomateriálních studiích a zdůrazňuje důležitost použití více typů buněk pro dosažení komplexního pochopení reakce imunitního systému na biomateriály.

Abstract:

The importance of biomaterials in biomedical application grows, they are used widely from drug carriers to tissue engineering. Understanding of the properties and interactions of these biomaterials with biological systems is essential for their successful utilization. One of the critical aspects of this issue is the evaluation of the immune system response, which has been recently the subject of intensive research. The reaction of immune system is assessed by in vitro and in vivo methods, but before proceeding to in vivo testing, testing of cytotoxicity and immuno-compatibility must take place. This is done by using in vitro methods, which provide convenient and cost-effective means of studying the immune system’s reaction to biomaterials. Therefore, this work focuses on the description of this reaction and the in vitro methods used to research the immune response to biomaterials on both established and less common cell populations. Specifically, it describes the potential of macrophages, neutrophils, dendritic cells, and lymphocytes for immuno-compatibility testing. In addition, this work explores the use of cocultures in biomaterial studies and highlights the importance of using multiple cell types to achieve a comprehensive understanding of the immune system’s response to biomaterials.
