Microbial amonium ion elimination from mine waters

Abstract:

V této bakalářské práci se věnuji biologickému odstraňování amonných iontů – procesu nitrifikace/denitrifikace. Náplní bakalářské práce bylo potvrzení průběhu nitrifikace a denitrifikace v retenční nádrži v Litvínově. Byly provedeny laboratorní testy vlivu teploty a množství biomasy na průběh nitrifikace a také monitorování hladiny biomasy během procesu nitrifikace. Byl také sledován vliv kyslíku na proces, jednalo se o limitaci procesu nitrifikace kyslíkem. Závěrem byl proveden experiment na prokázání přítomnosti nitrifikačních bakterií a identifikace jednotlivých druhů bakterií obsažených v používaném aktivovaném kalu. Teoretická část popisuje dusík, jeho koloběh a také rizika výskytu ve vodách. Další část je zaměřena na biologické odstraňování amonných iontů – proces nitrifikace/denitrifikace a faktory ovlivňující tyto procesy. Jsou zde popsány nitrifikační a denitrifikační bakterie a různé biologické postupy odstraňování dusíku. Experimentální výsledky zahrnují vliv teploty a množství biomasy na průběh nitrifikace, studium limitace kyslíkem a průkaz nitrifikačních bakterií. Proces nitrifikace probíhal při všech testovaných teplotách (15, 20, 25 a 30 °C) a koncentrace amonných iontů prokazatelně klesla ve všech testovaných případech již po 48 hodinách. S klesající sušinou biomasy nitrifikačního kalu docházelo rovněž k poklesu rychlosti úbytku amonných iontů. Při teplotě 25 °C a výchozí koncentraci sušiny AK 0,6 g/l došlo v prvních 24 hodinách k zhruba stejnému nárůstu biomasy jako u výchozí koncentrace 0,3 g/l sušiny AK. S klesající teplotou docházelo ke zpomalování nárůstu biomasy. Koncentrace kyslíku 2 mg/l je dostatečná pro průběh nitrifikace. Koncentrace biomasy se zvyšuje i při nižších hodnotách rozpuštěného kyslíku. Přítomnost nitrifikačních bakterií v aktivovaném kalu byla prokázána.

Abstract:

The bachelor thesis deals with biological elimination of ammonium ion – process of nitrification and denitrification. The assignment was to confirm running process of nitrification and denitrification in retention pond in Litvínov. Laboratory test were carried out to determine influence of temperature and biomass amount on process of nitrification. The level of biomass amount was also monitored during the process of nitrification. The limitation of nitrification by oxygen was observed. Finally the experiment was carried out to confirm the presence of nitrification bacteria species in used activated sludge followed by their identification. In theory the nitrogen, nitrogen cycle and risks of nitrogen in water are described. The next part is focused on biological elimination of ammonia ion also called process of nitrification and denitrification and factors affecting the mentioned process. Nitrification/denitrification bacteria species and different biological elimination techniques are described. The process of nitrification was going on under all tested temperatures (15, 20, 25 a 30 °C) and concentration of ammonia ion was decreasing in all cases already after 48 hours. With decreasing biomass dry matter in used activated sludge the ammonium elimination speed also decreased. With starting concentration of biomass 0,6 g of dry matter per liter and temperature 25 °C the growth of biomass in the first 24 hours was almost the same as in the case with starting concentration of biomass 0,3 g of dry matter per liter and same temperature. With decreasing temperature the growth of biomass also decreased. The oxygen concentration 2 mg/l is sufficient for process of nitrification. The growth of biomass was also confirmed under lower dissolved oxygen concentration. The presence of nitrification bacteria species in used activated sludge was proved.
