The use of bacteriophages for treatment and prevention of infectious diseases

Abstract:

Antibiotic resistance is one of the major problems of modern medicine and the treatment of bacterial diseases is becoming increasingly difficult. One promising solution is the therapeutic use of bacteriophages - phage therapy. This thesis includes information on clinically important pathogens against which phage therapy has considerable potential, as well as properties and benefits of bacteriophages that are suitable for this method of treatment. The main part of the thesis is focused on the practical experience in the usage of phage therapy. At first, it addresses experimental studies on animals, which include the effect of dose and form of preparation, method of administration, and timing on the results of treatment. The use of phage therapy in humans is described in specific cases from the initial studies at the beginning of the last century to the present. It also depicts the current state of phage therapy in selected countries and the availability of phage preparations on the market. Finally, it shows the therapeutic potential of phage lytic enzymes - endolysins.

Abstract:

Antibiotická rezistence je jedním z hlavních problémů moderní medicíny a léčba bakteriálních onemocnění je čím dál těžší. Jedním slibným řešením je terapeutické použití bakteriofágů - fágová terapie. V této práci jsou uvedeny klinicky významné patogeny, proti nimž má fágová terapie značný potenciál, a také vlastnosti a výhody bakteriofágů vhodných pro tento způsob léčby. Hlavní část práce se zaměřuje na praktické zkušenosti s použitím fágové terapie. Nejprve se věnuje experimentálním studiím na zvířatech, které zahrnují vliv dávky a formy preparátu, způsobu jeho podání a načasování na výsledky léčby. Použití fágové terapie u lidí je popsáno na jednotlivých připadech od počátečních studií na začátku minulého století až po současnost. Rovněž je uveden současný stav fágové terapie ve vybraných zemích a dostupnost fágových preparátů na trhu. Na závěr je zobrazen terapeutický potenciál fágových lytických enzymů - endolyzinů.
