Thermokinetic characterization of the griseofulvin drug

Abstract:

Předložená diplomová práce je zaměřena na zkoumání amorfního léčiva griseofulvinu, které se používá jako antimykotikum k léčbě plísňové infekce. Griseofulvin byl zkoumán řadou metod. Ke zkoumání krystalizace sloužila optická mikroskopie a diferenční skenovací kalorimetrie. Diferenční skenovací kalorimetrie byla taktéž použita ke zkoumání kinetiky krystalizace a následně pomocí kinetického modelu byla určena predikce chování griseofulvinu. Další použitá metoda v této práci byla termogravimetrie, pomocí které byl zkoumán teplotní rozklad griseofulvinu. Nakonec byl griseofulvin podroben disoluční zkoušce, která je pro farmaceutický průmysl velmi důležitá.

Abstract:

The presented diploma thesis is focused on the investigation of the amorphous drug griseofulvin, which is used as an antifungal drug for the treatment of fungal infection. Griseofulvin has been investigated by a number of methods. Optical microscopy and differential scanning calorimetry were used to investigate crystallization. Differential scanning calorimetry was also used to investigate the kinetics of crystallization, and then a kinetic model was used to predict the behavior of griseofulvin. Another method used in this work was thermogravimetry, which was used to investigate the thermal decomposition of griseofulvin. Finally, griseofulvin was subjected to a dissolution test, which is very important for the pharmaceutical industry.
