Antimicrobial Action of Colicins on Bacteria Producing Biogenic Amines

Abstract:

Biogenní aminy jsou obsaženy téměř ve všech potravinách rostlinného, ale zejména živočišného původu. Nejčastěji jsou produktem mikrobiálních dekarboxyláz. Výskyt biogenních aminů v potravinách může představovat závažné zdravotní riziko pro citlivé konzumenty. Redukce množství biogenních aminů v potravinách je otázkou snížení či likvidace dekarboxyláza pozitivní mikroflóry. Cílem této práce bylo otestovat potenciál použití gramnegativních bakteriocinů (koliciny, mikrociny) k redukci biogenních aminů v mase. Koliciny a mikrociny jsou antimikrobiálně působící proteiny na taxonomicky blízce příbuzné bakterie. Nejdříve byl sledován antimikrobiální účinek bakteriocinů na dekarboxyláza pozitivní bakterie izolované z masa bažantů. Z tohoto testování byly vybrány čtyři kmeny Escherichia coli s nejširším spektrem účinku, z nichž byly izolovány surové bakteriociny. Bylo zjištěno, že vzhledem k tomu, že gramnegativní bakteriociny neinhibují dostatečné množství dekarboxyláza pozitivních kmenů a jsou nestabilní při skladování v chladničce, nejsou ideálním prostředkem pro redukci biogenních aminů v mase.

Abstract:

Biogenic amines are found almost in all the food of plant, but especially of animal origin. Most often, they are the product of microbial decarboxylases. The occurrence of biogenic amines in food can represent a serious health risk to sensitive consumers. The quantity re-duction of biogenic amines in food is a matter of reducing or eliminating the decarboxylase and the positive microflora. The aim of this work was to test the potential of using gram-negative bacteriocins (colicins, microcins) to reduce biogenic amines in meat. Colicins and microcins are antimicrobially active proteins which affect taxonomically closely related bacteria. The first thing being done was monitoring the antimicrobial effect of bacteriocins on the decarboxylase and the positive bacteria from pheasant meat. From this testing, four strains of E. coli with the widest spectrum of effect were selected, from which the crude bacteriocins were isolated. It was found that since gram-negative bacteriocins do not inhibit sufficient amount of decarboxylase positive strains and are unstable when stored in the refrigerator, they do not serve as an ideal way of reducing biogenic amines in meat.
