Stanovení disociačních konstant antidepresiva Vortioxetine a léčiva Lesinuradu, k léčbě hyperurikémie spojené s dnou

Determination of dissociation constants of the antidepressant Vortioxetine and Lesinurade, for the treatment of hyperuricaemia associated with gout

Abstract:

Byly porovnány metody potenciometrické a spektrofotometrické pH-titrace u antidepresiva Vortioxetinu ke stanovení disociačních konstant. Vortioxetin je atypické antidepresivum, tj. modulátor a stimulátor serotoninu. Depresivními poruchami jsou chápány běžné psychické stavy, o nichž se domníváme, že jsou způsobeny nerovnováhou serotoninu a norepinefrinu, kromě mnoha dalších situačních, kognitivních a lékařských faktorů. Chemometrický přístup k nelineární regresi pH-spekter (REACTLAB, SQUAD84) a pH-titrace (ESAB) určil dvě překrývající se disociační konstanty. Mírně rozpustná neutrální báze LH hydrobromidu Vortioxetinu byla schopná protonace za vzniku dvou kationtů LH2+ a LH32+ rozpustných v čisté vodě. V rozmezí pH 5 až 10 lze spolehlivě odhadnout dvě disociační konstanty z malých změn ve spektrech při použité koncentraci 9.2 × 10-5 M Vortioxetinu. I když změna pH poněkud méně ovlivnila změny v chromoforu molekuly, byly odhadnuty dvě thermodynamické disociační konstanty pKTa1 = 7.22 a pKTa2 = 8.67 při 25 °C a pKTa1 = 7.27 a pKTa2 = 8.79 při 37 °C. Graf molárních absorpčních koeficientů různě protonovaných částic dle vlnové délky ukazuje, že spektrum částic LH2+ a LH se liší barvou, zatímco protonace chromoforu LH2+ na LH32+ má již menší vliv na chromofor molekuly Vortioxetinu hydrobromidu. Dvě thermodynamické disociační konstanty při použité koncentraci 3 × 10-4 M Vortioxetinu byly stanoveny regresní analýzou potenciometrických pH-titračních křivek pKTa1 = 7.08 a pKTa2 = 8.50 při 25 °C a pKTa1 = 7.33 a pKTa2 = 8.76 při 37 °C. Predikce disociačních konstant Vortioxetinu byla provedena programy MARVIN a ACD/Percepta a byly predikovány dvě disociační konstanty. Dále byly porovnány potenciometrické a spektrofotometrické pH-titrace Lesinuradu při stanovení tří disociačních konstant. Lesinurad je selektivní inhibitor reabsorpce kyseliny močové v rámci kombinace léků pro léčbu vysokých hladin kyseliny močové v krvi, nazývané hyperurikémie. Nelineární regrese pH spektra programy REACTLAB a SQUAD84, a pH-titrační křivky programem ESAB, stanovila tři postupné disociační konstanty. Bylo navrženo protonační schéma léčiva Lesinurad. Rozpustný anion L- byl protonován do rozpustných LH, LH2+ a LH32+ iontů v čisté vodě. Byly odhadnuty tři postupné thermodynamické disociační konstanty pKTa1 = 2.09, pKTa2 = 4.25, pKTa3 = 6.58 při 25 °C a pKTa1 = 1.96, pKTa2 = 4.16, pKTa3 = 6.32 při 37 °C, analýzou UV spekter. Graf molárních absorpčních koeficientů ukazuje, že spektrum LH2+ a LH se liší barvou, zatímco protonace chromoforu LH2+ na LH32+ má menší vliv na chromofory v molekule Lesinuradu. Tři thermodynamické disociační konstanty při koncentraci 1 × 10-4 mol. dm-3 Lesinuradu, byly stanoveny pomocí pH-potenciometrické analýzy pKTa1 = 2.39, pKTa2 = 3.47, pKTa3 = 6.17 při 25 °C a pKTa1 = 2.08, pKTa2 = 3.29, pKTa3 = 6.03 při 37 °C. Hodnoty enthalpie H0 (pKa1) = 19.19 kJ.mol-1, H0 (pKa2) = 13.29 kJ.mol-1, H0 (pKa3) = 38.39 kJ.mol-1, ukazují, že disociační proces je endothermní. Pozitivní hodnoty G0 (pKa1) = 11.93 kJ.mol-1, G0 (pKa2) = 24.26 kJ.mol-1, G0 (pKa3) = 36.56 kJ.mol-1 při 25 °C naznačují, že disociační proces není spontánní, což bylo potvrzeno hodnotou enthropie S0 (pKa1) = 24.37 J.mol-1, S0 (pKa2) = -36.79 J.mol-1, S0 (pKa3) = 2.79 J.mol-1. Tři disociační konstanty Lesinuradu a protonační lokality byly předpovězeny programem MARVIN a ACD/Percepta.

Abstract:

Potentiometric and spectrophotometric pH-titrations of the antidepressant Vortioxetine for dissociation constants determination were compared. Vortioxetine is an atypical antidepressant i.e. a serotonin modulator and stimulator. Depressive disorders are common mental health conditions thought to be caused by an imbalance in serotonin and norepinephrine in addition to multiple situational, cognitive, and medical factors. A chemometrics approach to nonlinear regression of the pH-spectra (REACTLAB, SQUAD84) and pH-titration (ESAB) determined two overlapping dissociation constants. A sparingly soluble neutral base LH of Vortioxetine hydrobromide was capable of protonation to form the still soluble two cations LH2+ and LH32+ in pure water. In the range of pH 5 to 10 two dissociation constants could be reliably estimated from small changes in spectra of 9.2 × 10-5 M Vortioxetine. Although the change of pH somewhat less affected changes in the chromophore, two thermodynamic dissociation constants were estimated pKTa1 = 7.22 and pKTa2 = 8.67 at 25 °C and pKTa1 = 7.27 and pKTa2 = 8.79 at 37 °C. The graph of molar absorption coefficients of variously protonated species according to wavelength shows that the spectrum of species LH2+ and LH vary in colour, while protonation of chromophore LH2+ to LH32+ has less influence on chromophores in Vortioxetine hydrobromide molecule. Two thermodynamic dissociation constants of 3 ×10-4 M Vortioxetine were determined by the regression analysis of potentiometric titration curves pKTa1 = 7.08 and pKTa2 = 8.50 at 25 °C and pKTa1 = 7.33 and pKTa2 = 8.76 at 37 °C. A prediction of the dissociation constants of Vortioxetine was carried out using the MARVIN and ACD/Percepta programs and only two dissociation constants were theoretically proposed. Potentiometric and spectrophotometric pH-titrations of the Lesinurad for three consecutive dissociation constants determination were compared. Lesinurad is a selective inhibitor of uric acid reabsorption as part of a combination of medicines to treat high levels of uric acid in blood, also called hyperuricemia. Nonlinear regression of the pH-spectra with REACTLAB and SQUAD84 and of the pH-titration curve with ESAB determined three multiple close dissociation constants. The protonation scheme of Lesinurad was suggested. A sparingly soluble anion L- of Lesinurad was protonated to the still soluble species LH, LH2+ and LH32+ in pure water. Three consecutive thermodynamic dissociation constants were estimated pKTa1 = 2.09, pKTa2 = 4.25, pKTa3 = 6.58 at 25 °C and pKTa1 = 1.96, pKTa2 = 4.16, pKTa3 = 6.32 at 37 °C by UV-metric spectra analysis. The graph of molar absorption coefficients shows that the spectrum of species LH2+ and LH vary in colour, while protonation of chromophore LH2+ to LH32+ has less influence on chromophores in the Lesinurad molecule. Three multiple thermodynamic dissociation constants of 1 × 10-4 mol. dm-3 Lesinurad were determined by the pH-metric analysis pKTa1 = 2.39, pKTa2 = 3.47, pKTa3 = 6.17 at 25 °C and pKTa1 = 2.08, pKTa2 = 3.29, pKTa3 = 6.03 at 37 °C. The values of enthalpy H0(pKa1) = 19.19 kJ.mol-1, H0(pKa2) = 13.29 kJ.mol-1, H0(pKa3) = 38.39 kJ.mol-1, show the dissociation process is endothermic. The positive values of G0(pKa1) = 11.93 kJ.mol-1, G0(pKa2) = 24.26 kJ.mol-1, G0(pKa3) = 37.56 kJ.mol-1 at 25 °C indicate that the dissociation process is not spontaneous, which was confirmed by the value of entropy S0(pKa1)= 24.37 J.mol-1, S0 (pKa2)= -36.79 J.mol-1, S0(pKa3)= 2.79 J.mol-1. Three macro-dissociation constants of Lesinurad and protonation locations were predicted by MARVIN and ACD/Percepta.
