Cardiomyogenesis in vitro with respect to HIF-1 alpha and p38 kinase roles

Abstract:

Kardiomyogeneze je proces tvorby srdečních buněk, který zahrnuje diferenciaci, proliferaci a migraci raných kardiomyocytů. Zralé kardiomyocyty lze rozpoznat jako spontánně bijící buněčnou kulturu, a také na základě exprese kardio-specifických genů. Do regulace procesu kardiomyogeneze je zapojeno velké množství faktorů, které ji mohou významně ovlivnit modulací různých signálních drah včetně klíčových HIF-1α a p38 MAPK. p38 MAPK zřejmě hraje roli v odpovědi na stresové podmínky v průběhu kardiomyogeneze in vitro i in vivo, avšak její funkce není doposud dostatečně objasněna. HIF-1α pomocí indukce transkripce mnoha genů reguluje množství pochodů během kardiomyogeneze. Hlavním cílem této práce bylo sledování průběhu kardiomyogeneze in vitro u dvou linií myších embryonálních kmenových buněk, jak kontrolních, tak deficientních na p38 MAPK a HIF-1α, diferencovaných pomocí dvou odlišných protokolů. Z výsledků vyplývá, že intenzivnější diferenciace bylo dosaženo pomocí standardního protokolu diferenciace adherentních buněk. Dále se zkoumal význam p38 MAPK a HIF-1α na průběh kardiomyogeneze in vitro. Zatímco delece HIF-1α působila spíše inhibičně, ztráta p38 MAPK neměla téměř žádný vliv. Poslední část výzkumu byla zaměřená na základní popis fyziologického stavu buněk v průběhu kardiomyogeneze in vitro. Pro tento účel byl vyhodnocen poměr exprese genů MHC-β ku MHC-α, ze kterého vyplývá, že kardiomyocyty diferencované in vitro se fenotypově podobají kardiomyocytům in vivo exponovaným stresovým podmínkám. Celkově tato práce přispěla k popisu průběhu kardiomyogeneze in vitro při optimálních kultivačních podmínkách pomocí kardio-specifických genů a osvětlila, jakou úlohu v tomto procesu hraje HIF-1α a p38 MAPK.

Abstract:

Cardiomyogenesis is the process of cardio cell formation which includes differentiation, proliferation and migration of early cardiomyocytes. Mature cardiomyocytes are characterized by the presence of spontaneously beating cells in culture and also by the expression of cardiac-specific genes. Cardiomyogenesis is regulated by a large number of factors that may significantly affect various signaling pathways, including HIF-1α and p38 MAPK. p38 MAPK is suggested to play a role in response to stress conditions during cardiomyogenesis in vitro and in vivo, but its function is still underexplored. HIF-1α is a key regulator of kardiomyogenesis responsible for the induction of transcription of many genes. The main aim of this thesis was to describe the cardiomyogenesis process in vitro in two different cell lines of murine embryonic stem cells, both control and deficient of p38 MAPK and HIF-1α, differentiated by two different protocols. The results show that intensive differentiation was achieved using a standard differentiation protocol employing adherent cells. Next, the role of p38 MAPK and HIF-1α during cardiomyogenesis in vitro was studied. The absence of HIF-1α inhibited the induction of cardiomyocyte differentiation. In contrast, the absence of p38 MAPK had almost no effect. The last part of this research was focused on the description of the physiological state of cells during in vitro cardiomyogenesis based on comparison of gene expression ratios of MHC-α to MHC-β. These results indicate that cardiomyocytes differentiated in vitro are phenotypically similar to in vivo cardiomyocytes exposed to stress conditions. In conclusion, this work contributed to description of cardiomyogenesis in vitro under optimal culture conditions by cardiac-specific genes and showed the role of HIF-1α and p38 MAPK in this process.
