Expression profiles of circulating miRNA as a potential biomarker of melanoma.

Abstract:

Tato bakalářská práce popisuje problematiku stanovení expresních profilů cirkulujících miRNA jako potenciálních biomarkerů melanomu. Teoretická část se věnuje procesu, který je charakteristický pro vznik a průběh kancerogeneze. Následuje kapitola o nádorovém onemocnění - maligním melanonu. Část teoretické bakalářské práce se také věnovala molekulám miRNA, významnosti tekuté biopsie a metodám pro stanovení expresních hladin miRNA metodou RT real-time PCR. Cílem praktické části bakalářské práce bylo zvládnutí metodiky izolace plazmy z plné krve a následné izolace RNA z plazmy pacientů chirurgicky léčených pro maligní melanom. Následně byla u vybraných pacientů stanovována exprese předoperačních a pooperačních hladin 23 molekul miRNA pomocí metody RT real-time PCR. Po následném vyhodnocení byl nalezen signifikantní vztah mezi předoperačními hladinami expresí 6 miRNA k celkové době přežití (OS) a signifikantní vztah 5 molekul miRNA k intervalu bez příznaků recidivy onemocnění (DFI). U předoperačních hladin 4 molekul miRNA nalezen signifikantní vztah jak k celkovému přežití, tak k intervalu bez příznaků recidivy onemocnění.

Abstract:

This bachelor thesis describes the problem of determining the expression profiles of circulating miRNAs as potential biomarkers of melanoma. The theoretical part is devoted to the process that is characteristic for the origin and progression of carcinogenesis. This is followed by a chapter focusing on malignant melanoma. The theoretical part of the bachelor thesis was also devoted to miRNA molecules, the importence of liquid biopsy and methods for determination of miRNA expression levels by RT real-time PCR. The aim of the practical part of the bachelor thesis was to master the methodology of isolation of plasma from whole blood and subsequent isolation of RNA from plasma of patients undergoing surgical treatment for malignant melanoma. Subsequently, the expression of preoperative and postoperative levels of 23 miRNA molecules was determined in selected patients using RT real-time PCR. After subsequent evaluation, a significant relationship between preoperative expression levels of 6 miRNAs and overall survival (OS) and a significant relationship of 5 miRNA molecules and disease-free interval (DFI) was found. In addition, a significant relationship for the preoperative levels of 4 miRNAs to both overall survival and symptom-free recurrence interval was found.
