Purification and characterization of potential adhesive protein from Candida albicans

Abstract:

Candida albicans is pathogenic yeast, which has a number of proteins on its cell wall that improve its ability to adhere onto the host mucous membrane. These proteins include glyceraldehyde 3 phosphate dehydrogenase. This primary glycolytic enzyme has many other functions. Because of its ability to adhere on host glycoproteins, it is one of the virulence factors. It has been shown, that in some organisms this enzyme can adhere to the saccharide part of the glycoproteins. It is possible that GAPDH from C. albicans also exhibit this feature. The aim of this diploma thesis was to purify GAPDH from C. albicans and characterize its features related to adhesive abilities. The conditions were optimized in order to prevent the formation of aggregates. Part of the thesis was focused on the separation of GAPDH from express cells, which created heterooligomers with GAPDH from C. albicans and were present in the solution after purification. The assumed ability of GAPDH to bind saccharides was examined by various methods.

Abstract:

Candida albicans je patogenní kvasinka, která na povrchu své buněčné stěny obsahuje mnoho proteinů, díky kterým je schopná adheze na sliznici hostitele. Mezi tyto proteiny patří i glyceraldehyd-3-fosfátdehydrogenáza. Tento primárně glykolytický enzym vykazuje mnoho dalších funkcí a mezi virulentní faktory ho zařazuje právě schopnost vázat hostitelské glykoproteiny. U některých organismů byla zjištěna schopnost vázat právě sacharidy glykoproteinů, a je možné, že tuto vlastnost vykazuje i GAPDH z C. albicans. Cílem diplomové práce bylo GAPDH z C. albicans purifikovat a charakterizovat vlastnosti související s jejími adhezivními schopnostmi. Byly optimalizovány podmínky, které zabránily tvorbě nerozpustných agregátů purifikované GAPDH. Část práce se zaobírala i separací GAPDH z expresního organismu, která tvořila heterooligomery s GAPDH z C. albicans a po purifikaci zůstávala v roztoku. Na základě předpokladu, že GAPDH je schopná vazby sacharidů, byly zkoumány i interakce GAPDH se sacharidovými strukturami.
