Interference of environmental contaminants with signalling pathways of poorly studied nuclear receptors

Abstract:

Endokrinní disrupce – schopnost látek narušit endokrinní systém organismů – byla v minulosti zkoumána převážně jako působení environmentálních polutantů skrze androgenní a estrogenní receptor. V poslední době je intenzivně studován také vliv xenobiotik na signální dráhy dalších nukleárních receptorů. Teoretická část bakalářské práce shrnuje dosavadní poznatky o zatím málo prostudovaných nukleárních receptorech (RAR, RXR, TR, PPAR, GR), způsobech endokrinní disrupce jejich signálních drah a možnostech testování těchto jevů in vivo a in vitro. V této části jsou také popsány některé známé důsledky působení xenobiotik skrze tyto nukleární receptory na organismy. V praktické části byla zkoumána schopnost syntetických hormonů dihydrotestosteronu a dexametazonu aktivovat signální dráhy androgenního a glukokortikoidního receptoru v buněčné linii MDA-kb2.

Abstract:

Endocrine disruption – the ability of substances to disrupt the endocrine system of organisms – has been investigated in the past mostly as effects of environmental pollutants on signalling pathway of androgen and estrogen receptor. Recently, the possible disruption of signalling pathways of other nuclear receptors has been studied intensively. The theoretical part of the thesis summarises the current knowledge about poorly studied nuclear receptors (RAR, RXR, TR, PPAR, GR), endocrine disruption of its signalling pathways, and methods of in vivo and in vitro testing. This part also describes some of the known effects of xenobiotics on poorly studied nuclear receptors. The ability of synthetic hormones dihydrotestosterone and dexamethasone to activate androgen and glucocorticoid receptor signalling pathways in the cell line MDA-kb2 were examined in the practical part of this thesis.
