Molecular mechanisms of zinc ions in prostate cancer

Abstract:

Prostatická tkáň je charakteristická a unikátní svým vztahem k zinku. Fyziologicky do-chází k vysoké míře akumulace tohoto kovu. Naopak, u nádorové tkáně dochází již od prvních stádií kancerogeneze k výraznému snížení koncentrace intracelulárního zinku. Cílem dizertační práce bylo nalézt odpověď na otázku, souvisí-li dysregulace zinečnatých iontů s pokročilostí onemocnění karcinomu prostaty a jakými mechanismy je tak činěno. Byl použit následující biologický materiál: prostatické buněčné linie, vzorky sér paci-entů s karcinomy. Byl využit MTT test a analýza buněčného růstu v reálném čase. Exprese genů byla analyzována pomocí real-time PCR a s využitím metod založených na imunodetekci. Na vícestupňovém modelu in vitro bylo prokázáno, že metastatické nádory vykazují nebývale vysokou mírou senzitivity k zinečnatým iontům. Ve shodě s tímto faktem jsou i poznatky získané studiem biologických vzorků pacientů s nádory: (1) hladina zinku v séru i v nádorové tkáni je nižší u pacientů oproti kontrolám (2) sérová hladina zinek-vázajícího pro-teinu metalothioneinu závisí na grade onemocnění a (3) vyšší hladiny metalothioneinu jsou aso-ciovány s horší prognózou pacientů. Na tomto se podílí několik mechanismů: byl prokázán zá-sadní vliv proteinu p53, dochází k inhibici apoptózy snížením exprese BAX, k nemožnosti zá-stavy buněčného cyklu a ke spuštění autofagie po působení stresorů různé povahy (chemotera-peutika, jiné oxidační stres-generující agens atd.). Zinečnaté ionty přispívají k ovlivnění přinejmenším tří charakteristických „schopností“ nádorů, tzv. „hallmarks of cancer“: odolávání buněčné smrti, deregulace energetického metabo-lismu a vyhýbání se supresorům růstu. Molekulární mechanismy spojené s rolí zinku jsou tak významnými kandidáty pro vývoj nových terapeutických strategií.

Abstract:

Prostatic tissue is characteristic in its relationship to zinc. A high degree of zinc accumulation physiologically occurs in the prostatic tissue. In contrast, tumour tissue is charac-teristic in a significant decrease of the intracellular zinc since the earliest stages of carcinogenesis. The aim of the thesis is to find an answer to the question, whether dysregula-tion of zinc ions is related to the advancement of the disease and what mechanisms are involved. Following biological materials were used: cell lines, sera from patients with tumours. MTT test and the analysis of cell growth in the real time were used to analyse the cytotoxic effect. Gene expression was analysed by real-time PCR and by immunoassays. It has been demonstrated that metastatic tumours exhibit an unusually high level of sensitivity to zinc ions. Consistent with this is also knowledge obtained by analysis of biolog-ical samples from patients with cancer: (1) level of zinc in the serum and tumour tissue is lower in patients versus controls (2) serum zinc-binding protein, metallothionein depends on the grade of the disease and (3) higher levels of metallothionein are associated with a worse prognosis. Several mechanisms are involved; distinctive effect of the p53 protein was determined, inhibi-tion of apoptosis by reducing the expression of BAX, inability to the cell cycle arrest and the triggering of the autophagy after stressors of different nature (chemotherapeutics, other oxidative stress-generating agent etc.) were observed. Zinc ions contribute to the alteration of at least three characteristic capabilities of tumours designated as „Hallmarks of cancer“: resisting cell death, deregulation of energy metabolism, and avoidance of growth suppressors. Molecular mechanisms associated with the role of zinc are thus important candidates for development of new therapeutic strategies.
